辅酶Q10影响急性心肌缺血再灌注损伤氧化应激和心功能的动物实验研究 被引量:4

摘要 目的探讨辅酶Q10影响急性心肌缺血再灌注(I/R)损伤氧化应激和心功能的动物实验研究。方法30只健康成年雄性Sprague-Dawley大鼠,按照随机数字表法分为3组:对照组、心肌I/R组和辅酶Q10组,每组各10只。对照组为假手术SD大鼠,进行0.9%氯化钠溶液肌肉注射;心肌I/R组为I/R模型大鼠,进行0.9%氯化钠溶液肌肉注射;辅酶Q10组为I/R模型大鼠,进行10 mg·kg^(-1)·d^(-1)辅酶Q10肌肉注射,均连续干预4周,剔除死亡及未成模大鼠,每组各有8只大鼠用于后续实验。通过大鼠酶联免疫吸附试验法试剂盒测定血清中肌酸激酶(CK)、乳酸脱氢酶(LDH)、超氧化物歧化酶(SOD)和还原型谷胱甘肽(GSH)活性以及丙二醛浓度。超声心动图和血流动力学分析急性心肌I/R损伤期间的心脏功能[射血分数(EF)、左心室收缩压(LVSP)、左心室舒张压(LVDP)、压力增速(+dP/dtmax)和压力降速(-dP/dtmax)]。通过心脏称重和TTC染色评估大鼠心肌肥厚和梗死。通过蛋白印迹分析自噬相关蛋白beclin-1、Atg5及LC-3A/B的蛋白表达。结果心肌I/R组较对照组CK和LDH的浓度升高,差异均有统计学意义(P<0.05);辅酶Q10组较心肌I/R组CK和LDH的浓度降低,差异均有统计学意义(P<0.05)。心肌I/R组较对照组SOD和GSH活性降低,丙二醛浓度升高,差异均有统计学意义(P<0.05);辅酶Q10组较心肌I/R组SOD和GSH活性升高,丙二醛浓度降低,差异均有统计学意义(P<0.05)。心肌I/R组较对照组LVDP、HW/BW比率和梗死面积升高,LVSP、+dp/dtmax、-dp/dtmax和EF等参数降低,差异均有统计学意义(P<0.05);辅酶Q10组较心肌I/R组LVDP、HW/BW比率和梗死面积降低,LVSP、+dp/dtmax、-dp/dtmax和EF等参数升高,差异均有统计学意义(P<0.05)。心肌I/R组较对照组beclin-1、Atg5和LC-3A/B的蛋白表达升高,差异均有统计学意义(P<0.05),辅酶Q10组较心肌I/R组beclin-1、Atg5蛋白表达升高,LC-3A/B蛋白表达降低,差异均有统计学意义(P<0.05)。心� Objective To investigate the effect of coenzyme Q10 on acute myocardial ischemia-reperfusion(I/R)injury,oxidative stress and cardiac function in animal experiments.Methods Thirty healthy adult male Sprague-Dawley rats were divided into three groups according to the random number table method:control group,myocardial I/R group and coenzyme Q10 group,10 rats in each group.The control group was sham-operated SD rats and received intramuscular injection of 0.9%sodium chloride solution;the myocardial I/R group was I/R model rats and received intramuscular injection of 0.9%sodium chloride solution;the coenzyme Q10 group was I/R model rats and received 10 mg·kg^(-1)·d^(-1)coenzyme Q10 was injected intramuscularly,and the rats were continuously intervened for 4 weeks.The dead and immature rats were eliminated,and there were 8 rats in each group.The activity of creatine kinase(CK),lactate dehydrogenase(LDH),superoxide dismutase(SOD)and reduced glutathione(GSH)activities and the concentration of malondialdehyde(MDA)were determined by rat enzyme-linked immunosorbent assay kit.Heart function[ejection fraction(EF),left ventricular systolic pressure(LVSP),left ventricular diastolic pressure(LVDP),pressure increase(+dP/dtmax)and pressure decrease(-dP/dtmax)]during acute myocardial I/R injury was analyzed by echocardiography and hemodynamics.The infarcted myocardial hypertrophy and infarction in rats were evaluated by heart weighing and TTC staining.The protein expression of autophagy-related proteins beclin-1,Atg5 and LC-3A/B were analyzed by Western blotting.Results The concentration of CK and LDH in the myocardial I/R group was higher than that in the control group,the difference was statistically significant(P<0.05);and the concentration of CK and LDH in the coenzyme Q10 group was lower than that in the myocardial I/R group,the difference was statistically significant(P<0.05).Compared with the control group,the activity of SOD and GSH was lower in the myocardial I/R group,and the concentration of MDA was higher,the differe
