摘要
目的:探讨瑞马唑仑对老年大鼠海马B淋巴细胞瘤-2(Bcl-2)、一氧化氮合酶(NOS)蛋白表达及神经元凋亡和认知功能的影响。方法:将39只亚急性衰老大鼠随机平分为模型组、咪达唑仑组和瑞马唑仑组。采用Morris水迷宫实验检测大鼠认知功能情况。检测血清中超氧化物歧化酶(SOD)活性与丙二醛(MDA)含量。采用TUNEL法检测CA1与CA3区神经元凋亡指数。采用Western blot检测海马组织Bcl-2、NOS的相对表达水平。结果:处理后第3、7天,咪达唑仑组和瑞马唑仑组逃避潜伏期高于模型组,且瑞马唑仑组低于咪达唑仑组(均P<0.05);咪达唑仑组和瑞马唑仑组穿越平台次数低于模型组,且瑞马唑仑组高于咪达唑仑组(均P<0.05)。处理后第3、7天,咪达唑仑组和瑞马唑仑组血清SOD活性高于模型组,MDA含量低于模型组(均P<0.05);瑞马唑仑组血清SOD活性高于咪达唑仑组,MDA含量低于咪达唑仑组(均P<0.05)。处理后第7天,咪达唑仑组和瑞马唑仑组海马CA1区和CA3区神经元凋亡指数低于模型组,且瑞马唑仑组低于咪达唑仑组(均P<0.05)。处理后第7天,咪达唑仑组和瑞马唑仑组海马组织Bcl-2、NOS蛋白相对表达水平高于模型组,且瑞马唑仑组高于咪达唑仑组(均P<0.05)。结论:瑞马唑仑能促进老年大鼠海马Bcl-2、NOS蛋白的表达,抑制神经元凋亡,对大鼠的认知功能影响较小。
Objective:To investigate the effects of remazolam on the expression of B-lymphoma-2(Bcl-2)and nitric oxide synthase(NOS)proteins,neuronal apoptosis and cognitive function in hippocampus of elderly rats.Methods:39 subacute aging rats were randomly divided into model group,midazolam group and rimazolam group.Morris water maze test was used to detect the cognitive function of rats.The activity of superoxide dismutase(SOD)and the content of malondialdehyde(MDA)in serum were detected.The apoptosis index of neurons in CA1 and CA3 regions was detected by TUNEL method,and the relative expression of Bcl-2 and NOS proteins in hippocampus was measured by Western blot.Results:The escape latency of midazolam group and rimazolam group on the 3rd and 7th day was higher than that of model group,and that of rimazolam group was lower than that of midazolam group(all P<0.05).The times of crossing the platform in midazolam group and remazolam group was lower than that in model group,and that in remazolam group was higher than that in midazolam group(all P<0.05).On the 3rd and 7th day after treatment,the activity of serum SOD in midazolam group and remazolam group was higher than that in model group,and the content of serum MDA was lower than that in model group(all P<0.05);the activity of serum SOD in remazolam group was higher than that in midazolam group,and the content of serum MDA was lower than that in midazolam group(all P<0.05).The apoptosis index of hippocampal CA1 and CA3 neurons in midazolam group and rimazolam group on the 7th day was lower than that in model group,and that in rimazolam group was lower than that in midazolam group(all P<0.05).The relative expression levels of Bcl-2 and NOS proteins in hippocampus of midazolam group and rimazolam group on the 7th day were higher than those of model group,and those of rimazolam group were higher than those of midazolam group(all P<0.05).Conclusion:Remazolam can promote the expression of Bcl-2 and NOS proteins in hippocampus of aged rats,inhibit neuronal apoptosis,and have l
