狂犬病病毒G蛋白的克隆表达及其抗体胶体金检测试纸条的研制 被引量:1

摘要 狂犬病(Rabies)是一种由狂犬病病毒(RV)引起的急性人兽共患传染病,一旦发病,死亡率几乎为100%。为快速检测狂犬病病毒IgG抗体,本研究首先克隆表达并纯化狂犬病病毒G蛋白,应用G蛋白作为抗原,G蛋白单克隆抗体制备C线,鼠抗犬IgG和鼠抗猫IgG标记T线,经条件优化后,建立了一种能够用于快速检测动物狂犬病病毒血清抗体水平的胶体金免疫试纸条。该试纸条具有快速、特异性、敏感性和重复性、稳定性较好的特点和优势,可对接种狂犬病疫苗动物的免疫接种效果进行快速定性评估,以消除免疫不确定动物带来的潜在威胁。 Rabies is an zoonotic disease caused by Rabies virus(RV).Once the disease occurs,the mortality rate is almost 100%.In order to rapidly detect RV IgG antibodies,the G gene was cloned and G protein was expressed and purifi ed for preparation of c-line,mouse anti-dog IgG and mouse anti-cat IgG were used to label T-line.After optimization of reaction conditions,a colloidal gold immunoassay strip was developed for rapid detection of serum antibody level of animal rabies virus infection.The test strip showed the advantages of rapidity,specifi city,sensitivity,repeatability,and stability.Therefore,it could be used for qualitative and rapid evaluation of the antibody responses to vaccinated animals to reduce the potential threat to animals with uncertain immunity.
