摘要
为了预防和应对新型鸭呼肠孤病毒引起的综合疾病,本研究利用分离到的新型鸭呼肠孤病毒,制备了具有良好中和活性的高效价IgY。中和试验结果显示:三免后第14天抗体滴度达到峰值,而后趋于平稳(>26),且添加CpG和IL-2分子佐剂的组别提取的IgY比未添加分子佐剂的组别中和滴度提高约3倍;将抗体注入SPF鸡胚进行攻毒保护试验,结果显示中和组的保护率最高,预防组的略高于治疗组的,且CpG组所提取的IgY预防效果(83%)和治疗效果(75%)最佳。本研究提取的高效价IgY能应用到临床防控中,可有效预防及治疗新型鸭呼肠孤病毒病。
To prevent and cure the comprehensive diseases caused by the novel duck reovirus,the high-titer egg yolk antibody IgY with good neutralizing activity was prepared with the isolated novel duck reovirus and purified in this study.The neutralization test results showed that the antibody titer peaked on the 14 th day after the third immunization,and then was stable above 26.The neutralization potency of the IgY extracted from the group with Cp G or IL-2 immune enhancer was almost 3 times higher than that from the group without molecular adjuvants.SPF chicken embryos were used for evaluating protective efficacy among them.The test results showed that the protection rate of the neutralization group is the highest,that of the prevention group is slightly higher than that of the treatment group;the IgY extracted from the Cp G group exhibited the best preventive rate(83%) and therapeutic rate(75%).The high titer IgY extracted in this study can be applied in clinical practice to effectively prevent and treat novel duck reovirus disease.
