摘要
以有机溶剂萃取制备的儿茶素粗品为材料,进行大孔吸附树脂柱色谱法制备高纯儿茶素(儿茶素含量≥95%)的研究。选用12种树脂为吸附剂,应用HPLC对吸附过程中儿茶素含量与组分的变化做了系统研究。结果表明:以脱咖啡因小叶种儿茶素粗品为原料,AB-8吸附树脂为柱填充料,收集过柱溶液一定区段内馏份,可以制得高纯儿茶素样品。产品制率为65%,纯度≥95%,EGCG含量大于55%,咖啡因<0.2%。
By using the crude catechin products produced by organic solvent extraction as raw material, the feasibility of preparing the high content catechins (content≥95%) with macroporous adsorption resin by column chromatography was studied. 12 kinds of resins were chosed as adsorbents, and the variations of catechins content and components during the process of static and dynamic adsorption were systematically tested by HPLC. Results showed that by using decaffeinated small leaf variety catechins as raw material, when a certain fraction collected through AB-8 chromatography, and concentrated under vacuum and freeze-dried, the high content catechins would be prepared. The recovery rate was 65%, the purity≥95%, EGCG>55% and caffeine <0.2%.
