摘要
[目的]探讨LncRNA GCMA在近远端胃癌组织中的表达及对胃癌细胞的凋亡作用。[方法]检测近远端胃癌组织和癌旁正常组织LncRNA GCMA的表达;用空白质粒、LncRNA GCMA抑制剂、过表达的LncRNA GCMA质粒转染胃癌AGS细胞,RT-PCR法、TUNEL染色法检测胃癌细胞凋亡率,Western Blot法检测上皮间质转化(EMT)相关分子ZO-1、E-cadherin、β-catenin和vimentin的表达。[结果]LncRNA GCMA在胃癌组织中高表达,近端胃癌组织中LncRNA GCMA表达量(5.52±1.56)高于远端(4.63±1.32)(P<0.05);近端胃癌病理分期、淋巴结转移与LncRNA GCMA表达呈正相关(P<0.05),远端胃癌淋巴结转移与LncRNA GCMA表达呈正相关(P<0.05)。分别下调和上调LncRNA GCMA,癌细胞凋亡率分别为(6.22±1.06)%、(3.32±0.77)%,高于对照组(8.63±1.41)%;下调LncRNA GCMA,可促进上皮细胞标志物ZO-1、E-cadherin的表达,抑制间质标志物β-catenin和vimentin的表达。[结论]LncRNA GCMA的表达与近远端胃癌细胞转移呈正相关关系,下调LncRNA GCMA可抑制细胞EMT过程,进而促进胃癌细胞凋亡。
[Objective]To investigate the expression of LncRNA GCMA in proximal and distal gastric cancer tissues and its apoptosis effect on gastric cancer cells.[Method]The expression of LncRNA GCMA in proximal and distal gastric cancer tissues and adjacent normal tissues was detected.Gastric cancer AGS cells were transfected with blank plasmid,LncRNA GCMA inhibitor,and overexpressed LncRNA GCMA plasmid.The apoptosis rate of gastric cancer cells was detected by RT-PCR and TUNEL staining.The expressions of ZO-1,E-cadherin,β-catenin and vimentin were detected by Western Blot.[Result]LncRNA GCMA was highly expressed in gastric cancer tissues,and the expression level of LncRNA GCMA was(5.52±1.56)in proximal gastric cancer tissues,which was higher than(4.63±1.32)in distal gastric cancer tissues(P<0.05).The pathological stage and lymph node metastasis rate of proximal gastric cancer were positively correlated with the expression of LncRNA GCMA(P<0.05),and lymph node metastasis rate of distal gastric cancer was positively correlated with the expression of LncRNA GCMA(P<0.05).After downregulation and upregulation of LncRNA GCMA,the apoptosis rates of cancer cells were(6.22±1.06)%and(3.32±0.77)%,respectively,which were higher than(8.63±1.41)%in control group;Down regulation of LncRNA GCMA could promote the expression of epithelial markers ZO-1 and E-cadherin,and inhibit the expression ofβ-catenin and vimentin.[Conclusion]LncRNA GCMA is positively correlated with metastasis of gastric cancer cells,and the down regulation of LncRNA GCMA can inhibit EMT process,thus promoting apoptosis of gastric cancer cells.
