摘要
目的检测乳腺癌组织中长链非编码RNA肝癌高表达转录本(lncRNA Hulc)、微小RNA-9-5p(miR-9-5p)的表达情况,探讨两者对乳腺癌淋巴结转移的预测价值。方法收集2017年1月至2019年1月贺州市人民医院诊治的80例乳腺癌患者的乳腺癌组织及其癌旁组织,应用实时荧光定量PCR检测乳腺癌组织和癌旁组织中lncRNA Hulc、miR-9-5p的表达水平;分析lncRNA Hulc、miR-9-5p的表达与乳腺癌患者临床病理参数的关系;采用Pearson相关性分析lncRNA Hulc与miR-9-5p表达的相关性,采用受试者工作特征(ROC)曲线分析lncRNA Hulc和miR-9-5p对乳腺癌淋巴结转移的预测价值。结果乳腺癌组织中的lncRNA Hulc表达水平高于癌旁组织,miR-9-5p表达水平低于癌旁组织,差异均有统计学意义(P<0.05);T3+T4期、Ⅲ+Ⅳ期及有淋巴结转移乳腺癌患者的lncRNA Hulc表达水平分别高于T1+T2、Ⅰ+Ⅱ期及无淋巴结转移者,Ⅲ+Ⅳ期、有淋巴结转移乳腺癌患者的miR-9-5p表达水平分别低于Ⅰ+Ⅱ期、无淋巴结转移者,差异均有统计学意义(P<0.05)。乳腺癌组织中lncRNA Hulc与miR-9-5p的表达呈负相关(r=-0.472,P=0.003)。lncRNA Hulc和miR-9-5p对乳腺癌淋巴结转移均有一定的预测价值,ROC曲线下面积均>0.7。二者联合应用的预测价值更高,敏感度和特异度分别为0.818、0.809。结论lncRNA Hulc在乳腺癌组织中表达增加,miR-9-5p在乳腺癌组织中表达下调,lncRNA Hulc和miR-9-5p的表达均与乳腺癌患者的不良病理参数相关,且对淋巴结转移有一定的预测价值。lncRNA Hulc、miR-9-5p可能是乳腺癌的治疗靶标。
Objective To detect the expression of long non-coding RNA highly upregulated in liver cancer(lncRNA Hulc)and microRNA-9-5p(miR-9-5p)in breast cancer tissues,and to explore the predictive value of both for lymph node metastasis of breast cancer.Methods 80 cases of breast cancer tissues and their adjacent tissues were collected from the patient with breast cancer who were treated in Hezhou people's hospital from January 2017 to January 2019,and quantitative real-time PCR was used to detect expression levels of lncRNA Hulc and miR-9-5p in breast cancer and adjacent tissues.The relationship between lncRNA Hulc,miR-9-5p expression and clinicopathological parameters of patients with breast cancer were analyzed.Pearson correlation was used to analyze the expression of lncRNA Hulc and miR-9-5p,and receiver operation characteristic(ROC)curve was used to analyze the predictive value of lncRNA Hulc and miR-9-5p in lymph node metastasis of breast cancer.Results The expression level of lncRNA Hulc in breast cancer was higher than that in the adjacent tissues,and the expression level of miR-9-5p was lower than that in the adjacent tissues,the difference was statistically significant(P<0.05).The expression of lncRNA Hulc in patient with stage T3+T4,stageⅢ+Ⅳor with lymph node metastasis was significantly higher than that in patient with stage T1+T2,stageⅠ+Ⅱor without lymph node metastasis,the expression level of miR-9-5p in patients with stageⅢ+Ⅳor with lymph node metastasis was lower than that in patients with stageⅠ+Ⅱor without lymph node metastasis,the difference was statistically significant(P<0.05).There was a negative correlation between lncRNA Hulc and miR-9-5p expression in breast cancer tissues(r=-0.472,P=0.003).LncRNA Hulc and miR-9-5p had a certain predictive value for lymph node metastasis of breast cancer,and the area under ROC curve was greater than 0.7.The prediction value of the combination of the two was higher,and the sensitivity and specificity were 0.818 and 0.809 respectively.Conclusions The
