梧州霉豆腐渣中微生物群落多样性及食用安全性分析 被引量:1

摘要 目的研究梧州霉豆腐渣微生物群落多样性和有害真菌毒素残留量,验证其食用安全性。方法先提取霉豆腐渣样品的总DNA,利用高通量测序技术,通过16s rDNA和真菌ITS区域进行PCR扩增及Miseq测序分析,然后再利用超高效液相色谱-质谱法同时测定样品中的黄曲霉毒素、赭曲霉毒素、桔青霉素、脱氧雪腐镰刀菌烯醇、玉米赤霉烯酮、T-2毒素、伏马毒素等16种真菌毒素残留。结果高通量测序获得细菌Tag数目51821个,OTU(operational taxonomic units)数目57个,考克氏菌属(Kocuria)占比25.90%、肠球菌属(Enterococcus)占比18.73%、坂崎肠杆菌属(Cronobacter)占比14.78%、皮生球菌属(Dermacoccus)占比9.27%;真菌Tag数目56741个,OTUs数目6个,主要为链孢霉属(Neurospora),占比达到98.17%;其常见16种真菌毒素残留量未检出。结论初步推测梧州霉豆腐渣特征“红毛”为链孢霉(Neurospora),但发现有影响其食用安全性的腐生菌及有害坂崎肠杆菌等,需要进一步跟踪研究明确其来源、具体含量及相应控制方法,以确保其食用安全性。 Objective To study the microbial community diversity and harmful mycotoxin residues of Wuzhou moldy tofu residue,and verify its edible safety.Methods The total DNA of moldy tofu residue samples was extracted,PCR amplification and miseq sequencing analysis were performed by 16 S rDNA and ITS region of fungi by high throughput sequencing technology,and then the residues of 16 kinds of mycotoxins such as aflatoxin,ochratoxin,citridin,deoxynienol,zearalenone,T-2 toxin and fumaroxin were determined by ultra performance liquid chromatography-mass spectrometry.Results 51821 bacterial tags and 57 OTU were obtained by high throughput sequencing.Kocuria accounted for 25.90%,Enterococcus 18.73%,Cronobacter 14.78% and Dermococcus 9.27%;the number of fungal tags was 56741,and the number of OTU was 6,mainly Neurospora,accounting for 98.17%;the residues of 16 kinds of common mycotoxins were not detected.Conclusion It is preliminarily speculated that the characteristic “red hair” of Wuzhou moldy tofu residue is Neurospora,but saprophytic bacteria and harmful Enterobacter sakazakii and other microorganisms affecting the use safety have been found.In the next step,we need to track and clarify its source,specific content and corresponding control methods to ensure its edible safety.
