摘要
检测乳制品中三聚氰胺含量最常用的方法是《GB/T 22388—2008原料乳与乳制品中三聚氰胺检测方法》中的高效液相色谱法。在使用高效液相色谱法进行分析时,流动相的种类和比例对于试验结果,尤其是对出峰时间、峰面积、峰型及回收率等效果影响极大,因此配置合适的流动相比例对于获得良好的图谱和结果极其重要。本文以流动相中离子对试剂和乙腈的比例为变量,探究当其比例为85:15、88:12、90:10、92:8、95:5时对于试验结果的影响规律,并找出在当前实验室环境下最适宜的流动相比例,以此提高检测结果的可靠性。
The most commonly used method to detect melamine content in dairy products is high performance liquid chromatography(HPLC)in GB/T22388—2008 method for detecting melamine in raw milk and dairy products.When using high performance liquid chromatography for analysis,the type and proportion of mobile phase have great influence on the test results,especially on the peak time,peak area,peak type and recovery rate.Therefore,it is extremely important to configure the appropriate mobile phase ratio for obtaining good atlas and results.In this paper,the ratio of ions to reagents and acetonitrile in mobile phase was taken as variable,and the influence of the ratio of 85:15,88:12,90:10,92:8 and 95:5 on the test results was explored,and the most suitable ratio of mobile phase in the current laboratory environment was found out,so as to improve the reliability of the test results.
