山楂通过肠道菌群治疗脂质代谢异常的实验研究 被引量:16

Shanzha(Fructus Crataegi)Attenuates Atherosclerosis in High-Fat Diet-Fed Apolipoprotein E-knockout Mice via Regulating Gut Flora

摘要 目的探讨山楂制剂调脂与肠道菌群的相关性。方法将100只SPF级载脂蛋白E基因敲除小鼠(C57BL/6J Apoe^(-/-)小鼠)随机分为5组,每组20只。分为空白对照组、动脉粥样硬化(atherosclerosis,AS)模型组、AS加抗生素对照组及AS加山楂加抗生素组、AS加山楂组。检测指标:检测小鼠血脂水平,包括低密度脂蛋白胆固醇(LDL-C)、甘油三酯(TG)、总胆固醇(TC)、高密度脂蛋白胆固醇(HDL-C);运用RT-PCR技术定量检测腹腔巨噬细胞清道夫受体A1(scavenger receptor class A type 1,SR-A1)、CD36mRNA的表达水平;运用Western-blot方法检测CD36,SR-A1的蛋白表达。结果比较空白对照组、AS模型组、AS加抗生素对照组及AS加山楂加抗生素组、AS加山楂组的血脂水平、CD36和SR-A1的mRNA水平和蛋白水平变化,结果表明:(1)通过与空白对照组比较,AS加山楂组有显著降脂效果;(2)通过与AS模型组比较,AS加抗生素对照组和AS加山楂加抗生素组及AS加山楂组均有降脂效果;(3)AS加山楂加抗生素组降脂水平高于AS加抗生素对照组;(4)AS加山楂降脂水平高于AS加山楂加抗生素组。结论山楂具有改善高胆碱饮食喂养造成动脉粥样硬化模型小鼠血脂的作用,山楂制剂调脂与肠道菌群相关。 Objective To explore the correlation between lipid-lowering of Shanzha(Fructus Crataegi)preparation and intestinal flora.Methods A total of 100 SPF apolipoprotein E gene knockout mice(C57 BL/6 J ApoE-/-mice)were randomly divided into 5 groups with 20 mice in each group:blank control group,atherosclerosis(AS)model group,AS plus antibiotic control group,AS plus Shanzha(Fructus Crataegi)plus antibiotic group and AS plus Shanzha(Fructus Crataegi)group.Indicators:the blood lipid levels of mice were measured,including low density lipoprotein cholesterol(LDL-C),triglyceride(TG),total cholesterol(TC)and high density lipoprotein cholesterol(HDL-C).RT-PCR was used to quantitatively detect the expression levels of scavenger receptor class a type 1(SR-A1)and CD36 mRNA in peritoneal macrophages.The protein expressions of CD36 and SR-A1 were detected by Western Blot.Results The changes of blood lipid levels,CD36 and SR-A1 mRNA and protein levels in blank control group,AS model group,AS plus antibiotic control group,AS plus Shanzha(Fructus Crataegi)plus antibiotic group and AS plus Shanzha(Fructus Crataegi)group were compared.1.Compared with blank control group,AS plus Shanzha(Fructus Crataegi)group had significant lipid-lowering effect.2.Compared with AS model group,AS plus antibiotic control group,AS plus Shanzha(Fructus Crataegi)plus antibiotic group and AS plus Shanzha(Fructus Crataegi)group had lipid-lowering effect.3.The lipid-lowering level of AS plus Shanzha(Fructus Crataegi)plus antibiotics group was higher than that of AS plus antibiotics control group.4.The lipid-lowering level of AS plus Shanzha(Fructus Crataegi)was higher than that of AS plus Shanzha(Fructus Crataegi)plus antibiotics group.Conclusions Our findings suggest that Shanzha(Fructus Crataegi)attenuates atherosclerosis in high-fat diet-fed ApoE;mice correlating with gut flora.
