摘要
目的:观察丹参、泽泻对非酒精性脂肪肝大鼠血管内皮功能的影响。方法:将60只SD大鼠随机分为6组,正常组、模型组、丹参组、泽泻组、丹泽组和东宝甘泰对照组,采用喂饲高脂饲料方法复制非酒精性脂肪肝模型,除正常组和模型组灌胃给予蒸馏水外,其余各给药组分别灌胃给予丹参水煎剂、泽泻水煎剂、丹参配伍泽泻水煎剂及东宝甘泰片治疗,连续8周,观察血清总胆固醇(TC)、甘油三酯(TG)、游离脂肪酸(FFA)、天门冬氨酸氨基转移酶(AST)、丙氨酸氨基转移酶(ALT)、超氧化物歧化酶(SOD)、丙二醛(MDA),内皮素(ET-1)、6-酮-前列腺素F1α6-Keto-PGF1α、血栓烷B2(TXB2)的含量或活性。结果:丹参、泽泻和丹参配伍泽泻水煎剂均能显著降低血清TC、TG、FFA、MDA、ALT、AST、SOD、ET、TXB2的含量或活性(P<0.01),同时升高6-Keto-PGF1α的含量(P<0.01)。结论:丹参、泽泻及丹参配伍泽泻水煎剂对非酒精性脂肪肝的防治作用,可能是通过改善氧化应激水平和血管内皮功能而实现。
Objective:To primarily discuss the effect of Salvia miltiorrhiza and Rrhizoma alismatis on the function of blood vessel endothelium in rats with non-alcoholic fatty liver disease.Methods:60 adult SD rats were randomly assigned into 6 groups(n=10 per group)named control group,model group,Salvia miltiorrhiza group,Rrhizoma alismatis group,Salvia miltiorrhiza and Rrhizoma alismatis group,and Dongbaogantai group.The rats’model of non-alcoholic fatty liver disease was duplicated by high-fat diet feeding.The control group and model group were given distilled water,other groups were given Salvia miltiorrhiza,Rrhizoma alismatis,Salvia miltiorrhiza and Rrhizoma alismatis,and Dongbaogantai.Salvia miltiorrhiza and Rrhizoma alismatis,as well as Dongbaogantai,was administrated in respectively designed groups,and corresponding indexes were detected 8 weeks later.Results:Salvia miltiorrhiza and Rrhizoma alismatis could significantly reduce serum concentration or activity of TC,TG,FFA,ALT,AST,SOD,MDA,ET,TXB2(P<0.01)and strengthening serum concentration of 6-Keto-PGF1α(P<0.01).Conclusion:Reducing the concentration of ET,and TXB2 and strengthening concentration of 6-Keto-PGF1αcould be viewed as mechanisms of Salvia miltiorrhiza and Rrhizoma alismatis on prevention and treatment of non-alcoholic fatty liver disease.
