摘要
目的探讨益糖康治疗2型糖尿病的可能作用机制。方法采用高脂饲料喂养结合一次性腹腔注射链脲佐菌素45 mg/kg制备2型糖尿病大鼠模型。将造模成功的32只SD大鼠随机分为模型组、益糖康组、二甲双胍组、益生菌组,每组8只,另设普通饲料喂养的SD大鼠8只为空白组。益糖康组给予益糖康生药56 g/(kg·d)灌胃,二甲双胍组给予盐酸二甲双胍片150 mg/(kg·d)灌胃,益生菌组给予益生菌冲剂150 mg/(kg·d)灌胃,空白组、模型组给予0.1 ml/10 g生理盐水灌胃,各组均每日1次。灌胃4周后,HE染色观察各组大鼠肠组织病理形态,测定血糖、胰岛素(INS)、糖化血红蛋白(HbAlc)、总胆固醇(TC)、甘油三酯(TG)、低密度脂蛋白胆固醇(LDL-C)、高密度脂蛋白胆固醇(HDL-C)水平;采用16S rRNA技术测定各组大鼠肠内容物的肠道菌群结构,进行α多样性指数分析、物种组成分析、LEfSe多级物种差异判别分析、COG功能注释,并对差异肠道菌群与血糖、血脂指标的相关性进行分析。结果病理结果显示,模型组、益生菌组均可见大量的肠绒毛顶端上皮细胞脱落;二甲双胍组偶见炎性细胞灶性浸润;益糖康组脱落的肠绒毛顶端上皮细胞较模型组明显减少。与空白组比较,模型组大鼠血糖、INS、HbAlc、TC、TG、LDL-C水平及coverage指数升高,HDL-C水平、shannon指数、chao指数降低,肠道菌群中放线菌门和变形菌门丰度升高,拟杆菌门丰度降低(P<0.05或P<0.01)。与模型组比较,二甲双胍组和益糖康组血糖、INS、HbAlc、TG、LDL-C水平降低,HDL-C水平、shannon指数升高,益生菌组血糖、coverage指数降低;二甲双胍组变形菌门丰度降低、疣微菌门丰度升高,益糖康组拟杆菌门、疣微菌门升高而变形菌门降低,益生菌组拟杆菌门、疣微菌门升高而放线菌门、其他菌门降低(P<0.05或P<0.01)。与二甲双胍组比较,益糖康组各指标差异均无统计学意义(P>0.05)。与�
Objective To explore the possible mechanism of Yitangkang(益糖康)in the treatment of type 2 diabetes mellitus(T2 DM).Methods The model of T2 DM was established by high-fat diet and one-time intraperitoneal injection of streptozotocin(STZ,45 mg/kg).The successfully-modeled 32 SD rats were randomly divided into model group,Yitangkang group,metformin group and probiotics group,with eight rats in each group,while 8 SD rats with ordinary diet were also set as blank group.The Yitangkang group was given 56 g/(kg·d)of crude Yitangkang by gavage;the metformin group was given 150 mg/(kg·d)of metformin hydrochloride tablets by gavage;the probiotics group was administered with 150 mg/(kg·d)of probiotics granules by gavage,while the blank group and model group were given 0.1 ml/10 g normal saline by gavage,once daily in all groups.After 4 weeks,the pathological morphology of the intestinal tissues were observed through HE staining,and the levels of blood glucose,insulin(INS),glycosylated hemoglobin(HbAlc),total cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-C)and high-density lipoprotein cholesterol(HDL-C)were measured;16 S rRNA technology was used to determine the intestinal flora structure of the intestinal content,andαdiversity index analysis,species composition analysis,the linear discriminant analysis effect size(LEfSe)of species differences,and the COG function annotation were performed;the correlations between the differential intestinal flora and blood sugar and blood lipids were analyzed.Results Pathological results showed that a large number of epithelial cells were shed from the apical intestinal villi in the model group and the probiotics group;inflammatory cell focal infiltration was occasionally seen in the metformin group;the shed epithelial cells of the apical intestinal villi of the Yitangkang group were significantly reduced compared to those of the model group.Compared to those of the blank group,the levels of blood glucose,INS,HbAlc,TC,TG and LDL-C and the coverage index o
