摘要
目的探讨滋肾活血方对血管性痴呆(Vascular dementia,VD)大鼠Nod样受体蛋白3(Nod-like receptor pyrin domain3,NLRP3)炎性小体通路的调控作用及海马神经血管单元(Neurova-scular Unit,NVU)的影响。方法选取SD大鼠60只进行VD造模,其中10只在手术过程只分离颈总动脉而不结扎作为假手术组,其余大鼠随机分为模型组,阳性药组,滋肾活血方低、中、高剂量组。阳性药组给予盐酸多哌奈齐,滋肾活血方低、中、高剂量组给予不同剂量的滋肾活血方,假手术组和模型组给予生理盐水,各组均灌胃给药。干预4周后,水迷宫法检测各组大鼠痴呆情况,试剂盒检测海马组织活性氧(Reactive oxygen species,ROS)、超氧化物岐化酶(Superoxide dismutase,SOD)、乳酸脱氢酶(lactate dehydrogenase,LDH)水平。Western blot法检测海马区神经元生长相关蛋白-43(Growth associated proteins-43,GAP-43)、突触素(Synaptophysin,SYN)、水通道蛋白4(Recombinant aquaporin 4AQP4)、NLRP3、凋亡相关斑点样蛋白(Apoptosis-related dot-like protein,ASC)、半胱天冬酶1(caspase-1)表达,ELISA法检测海马组织通路下游炎性因子白介素-18(Interleukin 18,IL-18)、白细胞介素-1β(Interleukin-1β,IL-1β)表达情况。结果模型组与滋肾活血方低、中剂量组大鼠逃避期均高于假手术组,跨平台数均低于假手术组,阳性药组,滋肾活血方低、中、高剂量组逃避期均低于模型组,跨平台数均高于模型组,滋肾活血方低、中剂量组逃避期均高于阳性药组,跨平台数均低于阳性药组(P<0.05)。模型组,阳性药组,滋肾活血方低、中剂量组ROS、LDH水平均高于假手术组,SOD水平均低于假手术组,阳性药组,滋肾活血方低、中、高剂量组ROS、LDH水平均低于模型组,SOD水平均高于模型组,滋肾活血方高剂量组ROS、LDH水平均低于阳性药组,SOD水平均高于阳性药组(P<0.05)。低、中剂量组ROS、SOD、LDH水平与阳性药组比较,差异无统计学意义(
Objective To explore the effect of Zishen Huoxue Recipe on Nod-like receptor pyrin domain 3(NLRP3)inflammatory pathway in vascular dementia(VD)rats and the Hippocampal neurovascular unit(NVU).Methods Sixty SD rats were selected for VD modeling.They were randomly divided into sham operation group,model group,positive drug group,low,medium and high dose groups.Ten of them were treated as the sham operation group by separating the common carotid artery without ligation during the operation.Model group,positive drug group,low,medium,and high dose groups were used to construct VD rat models.The positive drug group was given dopenezil hydrochloride,the low,medium,and high dose groups were given different doses of Zishen Huoxue Recipe,and the other groups were given normal saline,all were given by intragastric administration.After 4 weeks of intervention,the Morris water maze method was used to detect each group for dementia in rats,the kit was used to detect the levels of reactive oxygen species(ROS),superoxide dismutase(SOD)and lactate dehydrogenase(lactate)in hippocampus.dehydrogenase,(LDH)levels.Western blot detection of hippocampal neuron growth-related protein-43(Growth Associated Proteins-43,GAP-43),synaptophysin(SYN),aquaporin 4(Recombinant Aquaporin 4 AQP4),NLRP3,apoptosis-related spots Apoptosis-related Dot-like Protein ASC Antibody(ASC),caspase-1(caspase-1)expression.ELISA method was used to detect the expression of inflammatory factors Interleukin 18(IL-18),leukocyte Interleukin-1β(Interleukin-1β,IL-1β)of the hippocampal tissue.Results The escape period of rats in the model group and the low and medium doses of Zishen Huoxue Recipe was higher than that of the sham operation group,and the number of cross-platforms was lower than that of the sham operation group,the positive drug group,and the low,medium and high doses of Zishen Huoxue Recipe group.The escape period was lower than that of the model group,and the number of cross-platforms was higher than that of the model group.The escape period of the low a
