摘要
目的观察蠲痹历节清方对急性痛风性关节炎(AGA)大鼠NLRP3炎症体相关蛋白及白介素-1β(IL-1β)表达的影响,探讨蠲痹历节清方防治AGA大鼠可能存在的机制。方法将50只SPF级雄性SD大鼠随机分为空白组、造模组,造模组造模成功后随机分成模型组、蠲痹历节清方组、依托考昔组、异甘草素组,并予相应药物干预,模型组及空白对照组予等量生理盐水。观察蠲痹历节清方对大鼠步态、关节肿胀程度的影响,干预48 h后检测大鼠血尿酸及通过酶联免疫吸附测定(ELISA)检测大鼠滑膜NLRP3、凋亡相关斑点样蛋(ASC)、半胱氨酸蛋白酶-1(Caspase-1)及IL-1β蛋白表达水平。结果与空白组对照组比较,模型组中大鼠步态较差,关节肿胀明显,血清中血尿酸水平显著升高,滑膜组织中NLRP3、ASC、Caspase-1蛋白、IL-1β蛋白表达显著升高(P<0.01);与模型组比较,蠲痹历节清方组、依托考昔组、异甘草素组中大鼠步态、关节肿胀程度均有改善,且NLRP3、ASC、Caspase-1蛋白及IL-1β蛋白的表达均降低(P<0.05),且蠲痹历节清方组比依托考昔组更显著(P<0.05)。结论蠲痹历节清方可能通过抑制NLRP3炎症体信号通路减少NLRP3、ASC、Caspase-1蛋白表达,抑制IL-1β分化成熟,从而抑制AGA大鼠局部组织炎症。
Objective:To observe the effect of Juanbi Lijieqing formula on NLRP3(NACHT,LRR and PYD domain containing protein 3)inflammatory body related protein and interleukin-1βin rats with Acute Gouty Arthritis(AGA),and to explore the possible mechanism of Juanbi Lijieqing formula in preventing and treating AGA rats.Methods:50 SPF male SD rats were randomly divided into blank group and model group.After the model group were successfully established,they were randomly divided into model group,Juanbi Lijieqing formula group,etocoxibgroup and isoglycyrrhizin group,and given corresponding drug intervention.The model group and blank control group were given the same amount of physiological salt water.To observe the effect of Juanbi Lijieqing formula on gait and joint swelling of rats.After 48 hours of intervention,serum uric acid was detected,and synovial NLRP3,apoptosis associated speck like protein(ASC),Caspase-1 and IL-1βwere detected by Enzyme-linked Immunosorbent Assay(ELISA)expression level.Results:Compared with the blank group and the control group,the gait of rats in the model group was poor,the joint swelling was obvious,the level of serum uric acid was significantly increased,and the levels of NLRP3,ASC,Caspase-1 protein and IL-1βin synovium were significantly increased,but the protein expression was significantly increased(P<0.01);Compared with the model group,the gait and joint swelling of rats in Juanbi Lijieqing formula group,etocoxib group and isoglycyrrhizin group were improved,and NLRP3,ASC,Caspase-1 protein and IL-1βThe expression of protein decreased(P<0.05),and the Juanbi Lijieqing formula group was more significant than etocoxib group(P<0.05).Conclusion:Juanbi Lijieqing formula may reduce the expression of NLRP3,ASC,Caspase-1 protein and inhibit IL-1βby inhibiting NLRP3 inflammatory signal pathway differentiation and maturation,so as to inhibit local tissue inflammation in AGA rats.
