摘要 目的:建立蛇床子中蛇床子素的含量测定方法。方法:采用高效液相色谱法,色谱柱选择Agilent ZORBAX SB-C18色谱柱(250 mm×4.6 mm,5μm),流动相为甲醇-体积分数0.05%磷酸溶液(体积比70:30),流速为1.0 mL/min,检测波长为322 nm,柱温30℃。结果:蛇床子素的线性关系方程为Y=4.172×10^(4)X+1.912×10^(2),在12.5~200μg/mL范围内的峰面积与进样浓度线性关系良好(R^(2)=0.9996)。精密度、重复性及稳定性试验的相对标准偏差(RSD)分别为1.28%、1.32%、0.87%,平均加样回收率为99.36%,RSD为1.53%。蛇床子中蛇床子素的平均含量为5.58 mg/g。结论:建立的高效液相色谱法方法简便、准确、快速,重复性好,专属性好,可应用于蛇床子中蛇床子素的含量测定。 Objective:To establish a method for the determination of osthol in fructus cnidii.Methods:The HPLC was performed on Agilent ZORBAX SB-C18 column(250 mm×4.6 mm,5μm)with mobile phase consisted of methanol-0.05%phosphoric acid solution(70:30 by volume)at a flow rate of 1.0 mL/min.The detection wavelength was set at 322 nm.The column temperature was 30℃.Results:The linear relationship equation of osthol was y=4.172×10^(4)X+1.912×10^(2),the peak area in the range of 12.5-200μg/mL had a good linear relationship with the injection concentration(R^(2)=0.9996).The RSDS of precision,repeatability and stability tests were 1.28%,1.32%and 0.87%respectively.The average recovery was 99.36%,and the RSD was 1.53%.The average content of osthol in fructus cnidii was 5.58 mg/g.Conclusion:The established HPLC method is simple,accurate,rapid,reproducible and specific.It can be applied to the determination of osthol in fructus cnidii.
