摘要
本文建立了一种测定提取物中橘皮素含量的高效液相色谱(HPLC)方法。具体以Diamonsil C18色谱柱(250×4.6 mm,5μm)为固定相、甲醇-0.1%甲酸水溶液(70∶30,V/V)为流动相,设定流速为1.0 mL/min、柱温为30℃、进样体积为10μL,并以320 nm作为检测波长。结果显示橘皮素在2-80μg/mL范围内呈现出良好的线性关系(R=0.9994),平均加样回收率在99.45±1.44%范围内,相对标准偏差(RSD)值为1.45%(n=6)。该方法简便、准确、灵敏度高,且专属性强。
A high performance liquid chromatography(HPLC) method was developed for the determination of tangeretin in plant extracts.Diamonsil C18 column(250×4.6 mm,5 μm) was used as the stationary phase,methanol-0.1 % formic acid aqueous solution(70 ∶ 30,V/V) was used as the mobile phase,the flow rate was set at 1.0 mL/min,the column temperature was set at 30 ℃,the sample volume was set at 10 μL,and the detection wavelength was 320 nm.The results showed that a good linear relationship was found in the range of 2-80 μg/mL(R=0.9994).The average recovery was in the range of 99.45±1.44%,and the relative standard deviation(RSD) was 1.45 %(n=6).The method was simple,accurate,with high sensitivity and specificity.
