The relationship between the expression of lncRNA LINC00460 and ADAR1 and the prognosis of breast cancer and its diagnostic value

摘要 目的探讨长链非编码RNA(lncRNA)LINC00460和RNA特异性腺苷脱氨酶1(ADAR1)表达水平与乳腺癌预后的关系及其诊断价值。方法回顾性收集2019年5月至2020年5月同济大学附属第一妇婴保健院收治的86例乳腺癌患者的乳腺组织和癌旁组织。采用qRT-PCR法测定乳腺癌组织和癌旁组织中LINC00460和ADAR1的表达,同时分析LINC00460和ADAR1表达与乳腺癌患者临床病理的关系。Pearson相关性分析法分析LINC00460和ADAR1的相关性;Kaplan-Meier生存分析法分析LINC00460和ADAR1与乳腺癌患者预后的关系;Cox多因素回归法分析乳腺癌预后的影响因素。结果乳腺癌组织中LINC00460和ADAR1的相对表达量为4.13±1.24和5.07±1.62,均明显高于癌旁组织(0.91±0.27和1.03±0.39),差异均有统计学意义(P<0.05)。乳腺癌组织中LINC00460和ADAR1的表达与TNM分期、淋巴结转移、病理类型有关(P<0.05)。乳腺癌组织中LINC00460与ADAR1的表达呈正相关(r=0.63,P<0.001)。LINC00460低表达患者的生存率为91.30%,显著高于LINC00460高表达者(77.50%),ADAR1低表达患者的生存率为91.83%,显著高于ADAR1高表达者(74.29%),差异均有统计学意义(P<0.05)。LINC00460、ADAR1、TNM分期、病理类型、淋巴结转移是影响乳腺癌预后独立的危险因素。结论LncRNALINC00460和ADAR1在乳腺癌组织中高表达,与TNM分期、淋巴结转移、病理类型有相关性,是影响乳腺癌预后的独立危险因素。 Objective To investigate the relationship between long chain non encoding RNA(lncRNA)LINC00460 and RNA specific adenosine deaminase 1(ADAR1)expression and prognosis of breast cancer and its diagnostic value.Methods The breast tissue and adjacent tissues of 86 breast cancer patients admitted to Shanghai First Maternity and Infant Hospital,Tongji University School of Medicine from May 2019 to May 2020 were retrospectively collected.The qRT-PCR method was used to determine the expression of LINC00460 and ADAR1 in breast cancer tissues and adjacent tissues.The relationship between the expression of LINC00460 and ADAR1 and the clinicopathology of breast cancer patients was analyzed at the same time.The correlation between LINC00460 and ADAR1 was analyzed by Pearson's correlation.Kaplan-Meier survival to analyze the relationship between LINC00460 and ADAR1 and the prognosis of breast cancer patients.Cox multivariate regression analysis of factors affecting the prognosis of breast cancer.Results The relative expressions of LINC00460 and ADAR1 in breast cancer tissues were 4.13±1.24 and 5.07±1.62,which were significantly higher than those in adjacent tissues(0.91±0.27 and 1.03±0.39),the differences were statistically significant(P<0.05).The expressions of LINC00460 and ADAR1 in breast cancer tissues were related to TNM staging,lymph node metastasis,and pathological types(P<0.05).The expression of LINC00460 and ADAR1 in breast cancer tissues was positively correlated(r=0.63,P<0.001).The survival rate of low LINC00460 expression was 91.30%,which was significantly higher than that of high LINC00460 expression(77.50%),the survival rate of low ADAR1 expression was 91.83%,which was significantly higher than that of high ADAR1 expression(74.29%),the differences were statistically significant(P<0.05).LINC00460,ADAR1,TNM staging,pathological type,and lymph node metastasis are independent risk factors for the prognosis of breast cancer.Conclusion LncRNA LINC00460 and ADAR1 are highly expressed in breast cancer tissues and are
