胸膜肺炎放线杆菌在不同培养基中生长、生物膜形成及Apx毒素分泌能力的差异研究 被引量:4

摘要 为比较胸膜肺炎放线杆菌(APP)血清1型~12型菌株在不同培养基中的生长、生物膜形成以及Apx毒素分泌能力的差异,选用5种细菌培养基BHI、LB、MHB、PPLO和TSB,首先测定APP不同血清型菌株在5种培养基中的生长曲线,进而测定不同血清型菌株在5种培养基中的生物膜形成能力,同时测定这些菌株对20种抗菌药物的耐药性,最后测定APP血清1型菌株在5种培养基中添加CaCl_(2)对ApxⅠ毒素分泌的影响。结果显示,APP不同血清型菌株在BHI和TSB中生长较好,而在MHB、LB和PPLO生长较差;在不同培养基中培养24 h生物膜形成能力为MHB>BHI>TSB>LB>PPLO,血清3、4、7型和12型参考菌株生物膜形成能力较强;药敏结果显示,APP不同血清型菌株仅对万古霉素耐药;Apx毒素分泌结果显示,ApxⅠ在MHB、LB和PPLO中表达量显著高于BHI和TSB,而且培养基中添加5 mmol/L CaCl_(2)后ApxⅠ分泌量显著增加。研究结果可为分析APP不同血清型菌株在不同培养基中的生物学特性以及疫苗研制提供参考。 To compare the growth,biofilm formation,and Apx toxin secretion ability of Actinobacillus pleuropneumoniae(APP)serovars 1-12 strains in different culture media,five bacterial culture media BHI,LB,MHB,PPLO and TSB were used in this experiment.Firstly,the growth curves of different APP strains were determined in the 5 culture media.Then the biofilm formation ability of APP serovars 1-12 was determined in the 5 culture media.The susceptibility of APP strains to 20 antimicrobials was analysed.Finally,the effects of different culture media and CaCl_(2)addition on the secretion of ApxI toxin were determined in APP serovar 1 reference strain.The growth results showed that APP strains grew better in BHI and TSB than in MHB,LB and PPLO.Comparison on the biofilm formation ability in different media of APP strains at 24 h postinoculation was MHB>BHI>TSB>LB>PPLO,and serovars 3,4,7 and 12 reference strains had stronger biofilm formation ability.The antimicrobial susceptibility results showed that resistance to vancomycin was observed in different APP strains.The Apx toxin secretion results showed that the expression level of ApxI was significantly higher in MHB,LB and PPLO than in BHI and TSB.The secretion level of ApxⅠincreased significantly with addition of 5 mM CaCl_(2)in the media.The results will be helpful for analyzing the biological characters of APP strains with different serovars in selected culture media.
