摘要
目的探讨淋巴细胞活化基因3(LAG-3)在复发性中的作用及其与Th17/Treg细胞的关系。方法选取2018年3月至2019年8月新疆医科大学第一附属医院收治的35例复发性流产患者作为病例组,另选取32例同期正常早期妊娠自愿行人工流产的健康女性作为对照组。采集两组外周血,应用ELISA法检测血浆中Lag-3、IL-17和Treg的表达水平,Pearson法分析Lag-3与IL-17、Treg和Treg/Th17相关性。结果病例组外周血中Lag-3、Treg及Treg/Th17低于对照组,病例组IL-17高于对照组,差异有统计学意义(P<0.05),病例组中存在Treg/Th17失衡;研究组lag-3表达与Treg/Th17呈正相关(r=0.518,P<0.05)。结论复发性流产中Lag-3呈现异常表达状态,与Treg/Th17失衡及相关细胞因子改变有关,与Treg/Th17比率呈正相关,Lag-3有望成为临床判断复发性流产患者发生胚胎停育的特异性指标。
Objective To explore the role of lymphocyte activation gene 3(LAG-3)in recurrent spontaneous abortion and its relationship with Th17/Treg cells.Methods From March 2018 to August 2019,35 patients with recurrent spontaneous abortion admitted to the First Affiliated Hospital of Xinjiang Medical University were selected as the case group,and 32 cases healthy women of normal early pregnancy voluntary abortion during the same period were selected as the control group.Two sets of peripheral blood were collected,and the expression levels of Lag-3,IL-17 and Treg in plasma were detected by ELISA.The correlation of Lag-3 with IL-17,Treg and Treg/Th17 was analyzed by Pearson method.Results The peripheral blood Lag-3,Treg and Treg/Th17 in the case group were lower than the control group,the IL-17 in the case group was higher than the control group,and the differences were statistically significant(P<0.05),and there was an imbalance of Treg/Th17 in the case group.Study group lag-3 expression was positively correlated with Treg/Th17(r=0.518,P<0.05).Conclusion Lag-3 is abnormally expressed in recurrent spontaneous abortion,which is related to Treg/Th17 imbalance and related cytokine changes,and is positively correlated with Treg/Th17 ratio.Lag-3 is expected to become a specific indicator of embryonic stop in patients with recurrent spontaneous abortion in clinical judgment.
