摘要
目的研究朱砂肾毒性的量效关系与肾脏蓄积机理及针灸减毒的方法。方法将Hg^(2+)作用于HEK293,并通过MTT实验研究朱砂肾毒性的量效关系;将朱砂连续给予家兔,并设置针灸对照组,用原系荧光分析法检测家兔血液及肠离体灌流液与肾脏中Hg^(2+)的浓度。结果Hg^(2+)的肾细胞毒性存在明确的量效关系,朱砂混悬液实验组家兔的肠离体灌流液与肾脏中Hg^(2+)的含量均高于朱砂饱和溶液实验组,针灸实验组家兔的肠离体灌流液与肾脏中Hg^(2+)的含量均低于非针灸实验组。结论朱砂有很强的吸附性,而且附着在小肠内壁的朱砂微粒会导致吸收相延长,进而导致Hg^(2+)在肾脏的蓄积量增加;针灸可以减弱朱砂微粒的吸附作,进而减少Hg^(2+)在肾脏的蓄积,从而起到减毒作用。
Objective To study the dose-effect relationship between cinnabar administration and nephrotoxicity,its accumulation mechanism in kidneys and the method of reducing toxicity by acupuncture.Methods Hg^(2+) was applied to HEK293,and the dose-effect relationship between cinnabar administration and nephrotoxicity was studied by the MTT assay.Cinnabar was continuously administered to rabbits,and an acupuncture control group was set up.The concentrations of Hg^(2+) in blood,isolated intestinal perfusion fluid and kidneys of the experimental rabbits were detected by the original fluorimetry.Results There was a clear dose-effect relationship between Hg^(2+) and renal cytotoxicity.The contents of Hg^(2+) in both the isolated intestinal perfusion fluid and kidneys of rabbits in the cinnabar suspension experimental group,while the contents of Hg^(2+) in both the isolated intestinal perfusion fluid and kidneys of rabbits in the acupuncture experimental group were lower than those in the non-acupuncture experimental group.Conclusion The strong adsorptivity of cinnabar makes its microparticles attached to the inner wall of the small intestine,which extends its absorption phase and causes a higher accumulation of Hg^(2+)in kidneys.Acupuncture and moxibustion can weaken the adsorption of cinnabar particles,reduce the accumulation of Hg^(2+) in kidneys and its toxicity.
