摘要
为了探究热应激下大鼠脾脏组织形态学及TLR2和TLR4表达的变化情况,本研究通过建立不同处理的大鼠热应激模型,即38℃-单次热应激(6 h)、38℃-多次热应激(2 h/次,3次)、43℃-单次热应激(6 h)、43℃-多次热应激(2 h/次,3次),以室温条件下饲养的大鼠作对照,收集各组脾脏组织进行组织学和TLR2、TLR4 mRNA表达检测.HE染色结果发现,与对照组大鼠脾脏相比,各处理组脾脏白髓减少,红髓增加,并且随着温度的增加,脾窦严重萎缩,红细胞数明显减少;与单次热应激组相比,多次热应激组边缘区变宽.qRT-PCR检测显示,与对照组相比,TLR2和TLR4 mRNA在38℃处理组中表达上调,而在43℃处理组下调.免疫荧光染色发现,TLR2和TLR4蛋白均分布于各处理组红髓区、边缘区的巨噬细胞膜上;TLR2蛋白分布于对照组和38℃处理组的脾索;TLR4蛋白分布于对照组和38℃处理组白髓区的巨噬细胞膜上.由此可见,TLR2和TLR4基因可能参与调控热应激大鼠脾脏巨噬细胞的免疫应答过程:低热环境可通过增强免疫机能应对热应激对脾脏的损伤,而在高热环境下脾脏组织严重受损,其免疫功能被抑制或破坏.
In order to explore the changes of rat spleen tissue morphology and TLR2 and TLR4 expression under heat stress,this study established a heat stress model of rats with different treatments,including 38℃-single heat stress group(6 h),38℃-multiple heat stress group(2 h/time,three times),43℃-single heat stress group(43℃,6 h),43℃-multiple heat stress group(43℃,2 h/time,three times),with rats raised at room temperature as controls,and spleen tissues of each group were collected for histology and TLR2 and TLR4 mRNA expression detection.The results of HE staining showed that compared with the spleen of the control group,the white pulp of the spleen in each treatment group is decreased,and the red pulp is increased.With the increase of temperature,the spleen sinus is severely atrophied,and the number of red blood cells is significantly reduced.Compared with the single heat stress groups,the marginal area of the multiple heat stress group becomes wider.The qPT-PCR results indicated that compared with the control group,the expression of TLR2 and TLR4 mRNA is up-regulated in 38℃treatment group,and down-regulated in the 43°C treatment group.Immunofluorescence staining revealed that both TLR2 and TLR4 proteins are distributed on the macrophage membranes in the red pulp and marginal zone of each treatment group.TLR2 protein is distributed in the splenic cord of the control group and 38°C treatment group,and TLR4 protein is also distributed on the macrophage membranes in the white pulp zone of the control group and 38°C treatment group.It can be concluded that TLR2 and TLR4 genes may be involved in the regulation of immune response process of spleen macrophages in heatstressed rats.That is,a low-heat environment can respond to heat stress damage by enhancing immune function,while spleen tissue is severely affected in a high-heat environment for its immune function is suppressed or destroyed.
