摘要
补骨素合成酶(PS)是胡萝卜营养物质补骨素生物合成的关键酶。从‘黑田五寸’胡萝卜中克隆得到DcPS基因,对其进行了进化和在胡萝卜根发育过程中表达水平分析。结果表明,DcPS基因长1518 bp,编码氨基酸505 aa,属于细胞色素P450超级基因家族。其氨基酸序列含有10个明显的特征,其中含有1个跨膜区,为亲水性蛋白。进化分析表明,胡萝卜与毒胡萝卜的亲缘关系最近,在同一分枝上,但与中国的水芹和鸭儿芹的亲缘关系较远。基因表达分析显示,DcPS基因主要是在胡萝卜发芽后60~100 d表达最高。
Psoralen synthase(PS) is the key enzyme of psoralen biosynthesis. DcPS gene was cloned from carrot of ’Kuroda’, and analyzed its evolving and expressing level during the development of carrot roots. The results showed that the DcPS gene was 1 518 bp, encoded 505 amino acid residues, belonging to the cytochrome P450 supergene family. Its amino acid sequence contains 10 characteristics, including a transmembrane region, which is a hydrophilic protein. Phylogenetic analysis shows that the carrot is a close relative of the Thapsia garganica, and share the same branch, but DcPS gene of carrot is distant with Chinese Umbelliferae plants, Oenanthe javanica and Cryptotaenia japonica. Gene expression level showed that the DcPS highest expression was found in carrots 60-100 d after germination.
