摘要
目的:观察滋补脾阴方药对脾阴虚大鼠胃肠激素及胃肠免疫功能的影响。方法:将18只Wistar雄性大鼠随机分为正常对照组、模型组和滋补脾阴方药组,每组6只,除正常对照组外,其余各组采用饮食不节、过度劳倦、温里药伤阴的方法构建脾阴虚证大鼠模型,连续给予相应药物2周。采用ELISA法检测大鼠血清血管活性肠肽(vasoactive intestinal peptide,VIP)和胃组织分泌型免疫球蛋白A(secretory immunoglobulin A,sIgA)水平;免疫组化法检测空肠组织VIP表达。结果:与模型组比较,滋补脾阴方药组大鼠血清VIP水平明显降低(P<0.001),大鼠胃组织sIgA水平明显升高(P<0.001),大鼠空肠组织VIP水平明显降低(P<0.01)。结论:滋补脾阴方药可能是通过下调大鼠血清和空肠组织VIP水平,上调大鼠胃组织sIgA水平,达到治疗脾阴虚证的作用。
Objective:To explore the influence of formulas and medicines for nourishing spleen yin(FMNSP)on gastrointestinal hormones and gastrointestinal immune function in model rats with spleen yin deficiency syndrome.Methods:A total of 18 Wistar male rats were randomly divided into the normal control group,the model group and the FMNSP group,with 6 rats in each group.Except the normal control group,the other groups established the rat model of spleen yin deficiency syndrome by compound factor method(the combined application of inappropriate diet,excessive fatigue and consumption of body).After the model was successfully prepared,the corresponding medicines were given continuously for 2 weeks.The levels of serum vasoactive intestinal peptide(VIP)and gastric secretory immunoglobulin A(sIgA)were detected by ELISA.The expression of VIP in jejunum was detected by immunohistochemistry.Results:Compared with the spleen yin deficiency group,the level of serum VIP in the FMNSP group decreased significantly(P<0.001),while the level of sIgA in rats′gastric tissue increased significantly(P<0.001),and the level of VIP in jejunum decreased significantly(P<0.01).Conclusion:Formulas and medicines for nourishing spleen yin may treat spleen yin deficiency syndrome by down-regulating the level of VIP in rats′serum and jejunum and up-regulating the level of sIgA in rats′gastric tissues.
