摘要
本试验旨在探讨脂多糖(LPS)和松弛素(RLX)对产后KM小鼠子宫复旧过程中炎性细胞因子分泌的影响。将产后KM小鼠随机分为LPS组、RLX组和PBS组,分别用LPS、RLX和PBS进行子宫灌注,建立子宫复旧不良模型与正常子宫复旧模型。ELISA法检测小鼠血清内雌二醇、孕酮以及子宫组织中TNF-α和IL-1β的水平,同时对子宫组织进行H.E.染色并观察。结果显示,LPS组雌二醇含量和TNF-α含量均显著高于PBS组(P<0.05);RLX组雌二醇水平极显著高于PBS组(P<0.01),孕酮水平在复旧初期至中期显著高于PBS组(P<0.05),IL-1β含量显著低于PBS组(P<0.05)。光镜观察发现,LPS组主要出现炎性反应,恶露排出障碍、子宫内膜的更新迟缓以及子宫腺体的分泌增多等情况,RLX组主要表现为肌肉收缩不全,延缓子宫体的宫腔体积及子宫壁厚度回缩。结果表明,LPS和RLX进行子宫灌注均可导致产后子宫复旧不良,推测由于复旧进程受阻,雌激素分泌增加以期刺激子宫收缩加快复旧过程;在RLX组,雌激素可能促进IL-1β的表达下调。
This experiment aimed to examine the effects of lipopolysaccharide(LPS)and relaxin(RLX)on the secretion of inflammatory cytokines during the uterine involution of postpartum KM mouse.Mice were randomly divided into LPS group,RLX group and PBS group.LPS,RLX and PBS were used to perfuse into uterus of postpartum mice to establish two models of poor uterine involution and one normal model.The levels of estradiol and progesterone in serum,TNF-αand IL-1βin uterine tissue were detected by ELISA,and the uterine tissues were sectioned and stained with H.E.staining and observed with microscope.The results showed that the estradiol content and TNF-αcontent in LPS group were significantly higher than that in PBS group(P<0.05).Estradiol level in RLX group was significantly higher than that in PBS group(P<0.01),progesterone level was significantly higher than that in PBS group(P<0.05)in the early to middle stage of uterine involution,and IL-1βcontent was significantly lower than that in PBS group(P<0.05).Microscopy revealed that inflammatory reactions,obstruction of lochia discharge,slow renewal of endometrium and increased secretion of uterine glands were much common in LPS group.RLX mainly caused muscle contraction problems and affected the volume thickness of uterus.These results indicated that both LPS and RLX could lead to poor uterine involution,on the possibilities that estrogen secretion increased to stimulate uterine contraction and accelerate the involution process due to the obstruction of involution process,and that mechanism of estrogen of action on the RLX group may be related to down-regulation of IL-1βexpression.
