红曲茯苓片中多糖含量测定方法的比较研究 被引量:9

摘要 目的筛选一种适合红曲茯苓片中多糖的含量测定方法。方法采用水提醇沉法提取红曲茯苓片中的多糖,分别采用苯酚硫酸法、蒽酮硫酸法和DNS法测定多糖含量,对比不同方法得到数据之间的差异和准确性。结果苯酚硫酸法和蒽酮硫酸法分别在0.01~0.12 mg和0.04~0.14 mg质量范围内线性良好,DNS法在0.02~0.06 mg/mL浓度范围内线性良好;苯酚硫酸法测定样品多糖稳定性、重复性和加样回收率均优于其他方法;含量测定结果分别为7.350 g/100g、8.975 g/100g和8.664 g/100g,蒽酮硫酸法测得值最高。相比而言,苯酚硫酸法显色稳定,重现性好,回收率高,结果更合理可靠。结论红曲茯苓片多糖含量测定时可优先考虑苯酚硫酸法。 Objective To select a suitable method for the determination of polysaccharides in Hongqu Fuling Tablets.Methods The polysaccharides in Hongqu Fuling Tablets were extracted by water extraction and alcohol precipitation method. The content of polysaccharides was determined by phenol sulfuric acid method, anthrone sulfuric acid method and DNS method, respectively. The differences and accuracy of data obtained by different methods were compared Results The linear values of phenol sulfuric acid method and anthrone sulfuric acid method were in the range of 0.01~0.12 mg and 0.04~0.14 mg, respectively. The linear values of DNS method were in the range of 0.02 ~ 0.06 mg/mL. The stability, reproducibility and recovery rate of the sample polysaccharides determined by phenol sulfuric acid method were better than other methods. The content determination results were 7.350 g/100 g, 8.975 g/100 g and 8.664 g/100 g, respectively, and the highest value was obtained by anthrone sulfuric acid method. Conclusion Content determination of polysaccharide in Hongqu Fuling Tablets should give priority to phenol sulfuric acid method.
