摘要
目的:分析IL-15在胸腺上皮肿瘤(TETs)组织中的表达及其与患者临床病理特征和预后的关系,探讨其参与的调控网络。方法:运用GEPIA2数据和免疫组织化学技术分析IL-15在正常胸腺组织和TETs组织中的表达差异;基于TCGA TETs数据,探究IL-15的表达与患者临床病理特征和预后的关系,并利用免疫组织化学技术进一步分析IL-15与临床特征相关性;利用cBioPortal、DAVID工具进行TCGA TETs IL-15共表达基因GO和KEGG富集分析;基于STRING数据库分析IL-15蛋白-蛋白相互作用网络。结果:IL-15在TETs组织中表达显著增加,且与组织学分型、患者是否伴有重症肌无力相关。IL-15高表达患者总生存期和无进展生存期显著缩短,可作为TETs患者预后的独立危险因素。IL-15共表达基因主要富集于免疫分子、免疫细胞介导的免疫应答相关的生物学功能和信号通路。蛋白互作网络结果显示IL-15互作蛋白主要参与JAK/STAT信号通路。结论:IL-15参与TETs的发生、发展,在TETs免疫微环境发挥复杂而重要的功能,可作为临床诊断与预后的评价指标。
Objective:To analyze the expression of IL-15 in thymic epithelial tumors(TETs),explore its correlation with clinico pathological characteristics and prognosis of TETs patients,and its potential regulation network.Methods:Using GEPIA2 database and immunohistochemistry to test expression difference in normal thymus tissues and TETs tissues.Correlation between expression of IL-15 and clinicopathological features and prognosis of TETs patients was investigated by analyzing TCGA database.And association between IL-15 and clinical characteristics was further confirmed by immunohistochemistry.Based on the application of cBioPortal and DAVID tools to analyze the GO and KEGG cluster analysis of IL-15 co-expressed genes in TETs.IL-15 protein-protein interaction network analysis was performed using SRTING tool.Results:IL-15 was significantly higher in TETs tissues than that in normal thymus tissues,and IL-15 expression was associated with histologic diagnosis of TETs and myasthenia gravis.High expression of IL-15 showed significant shorter overall survival and progression free survival in TETs patients.Multivariate Cox regression analysis further indicated that IL-15 was an independent prognostic factor for survival in TETs.IL-15 co-expressed genes were mainly concentrated in the biological functions and signaling pathways related to immune molecules and immune cell mediated immune response.The protein-protein interaction network of IL-15 showed that it mainly involved in JAK/STAT signaling pathway.Conclusion:IL-15 may be involved in occurrence and development of TETs and plays a complicated and important role in immune microenvironment of TETs.It can be used as a reliable indicator of clinical diagnosis and prognosis.
