摘要
目的:本研究分析miR-223对呼吸衰竭患者Th1/Th2细胞功能及免疫功能的影响,以期为呼吸衰竭患者的临床治疗和生物靶标的选择方面提供更多参考资料。方法:抽取我院收治的40例呼吸衰竭患者为Ref组,40例同期健康体检者为Con组。采用荧光定量PCR实验测定miR-223的mRNA表达;采用PBMC法分离外周血单个核细胞,采用电穿孔顺式转染法将miR-223和对照组质粒转入核细胞;采用ELISA法测定miR-223对免疫功能的影响;采用流式细胞术测定miR-223对Th1/Th2细胞的影响;采用ELISA法测定miR-223对Th1/Th2细胞因子的影响。结果:miR-223 mRNA表达在Ref组中的水平相较Con组极显著上调(外周血和细胞)(P<0.01);IgA、IgM和IgG在Ref组中的水平相较Con组均极显著下调(P<0.01);Ref组的Th1/Th2细胞水平相较Con组均极显著下调(P<0.01);Th1类细胞因子中IL-2、IFN-γ和TNF-α在Ref组中的水平相较Con组均显著上调(P<0.05);Th2类细胞因子中IL-6、IL-10和IL-4在Ref组中的水平相较Con组均显著上调(P<0.05)。结论:miR-223可以下调呼吸衰竭患者的免疫球蛋白水平和Th1/Th2细胞水平,并且上调Th1/Th2细胞因子水平。
Objective:To investigate the effect of miR-223 on Th1/Th2 cell function and immune function in patients with respiratory failure,in addition to provide more reference for clinical treatment and selection of biological targets for patients with respiratory failure.Methods:Forty patients with respiratory failure admitted to our hospital were enrolled in the Ref group,and 40 healthy subjects in the same period were in the Con group.mRNA expression of miR-223 was determined by real-time PCR assay.Peripheral blood mononuclear cells were isolated by PBMC method.The miR-223 and control plasmids were transferred into nuclear cells by electroporation cis-transfection.The influence of miR-223 in immune function was determined by ELISA.The effect of miR-223 on Th1/Th2 cells was determined by flow cytometry.The effect of miR-223 on Th1/Th2 cytokines was determined by ELISA.Results:The level of miR-223 mRNA expression in the Ref group was significantly higher than that in the Con group(peripheral blood and cells)(P<0.01);the levels of IgA,IgM and IgG in the Ref group were significantly lower than those in the Con group(P<0.01);Th1/Th2 cell levels in Ref group were significantly down-regulated compared with Con group(P<0.01).The levels of IL-2,IFN-γand TNF-αof Th1 cytokines in the Ref group were significantly higher than those in the Con group significantly up-regulated(P<0.05).The levels of IL-6,IL-10 and IL-4 in the Th2 cytokines were significantly higher than those in the Con group(P<0.05).Conclusion:miR-223 can down-regulate immunoglobulin and Th1/Th2 cells in patients with respiratory failure and up-regulate Th1/Th2 cytokine levels.
