摘要
目的观察肾康灵对慢性肾脏病(CKD)小鼠肾功能和肾脏自噬水平的影响,探讨其对CKD肾损伤的保护机制。方法24只C56BL/6小鼠随机分为正常组、模型组和肾康灵组,每组8只。腺嘌呤诱导法复制CKD小鼠模型,肾康灵组予肾康灵药液(1.5 g/kg)灌胃,连续28 d。生化分析仪检测小鼠血尿素氮和血肌酐含量,HE染色、Masson染色和糖原PAS染色观察小鼠肾组织病理形态,透射电镜观察小鼠肾组织超微结构和自噬体变化,Western blot检测小鼠肾组织自噬相关蛋白p62、Beclin 1、Atg5和LC3B表达。结果与正常组比较,模型组小鼠肾脏萎缩,肾质量明显降低(P<0.01),血尿素氮和血肌酐含量显著增加(P<0.01),肾组织纤维化病变明显,Beclin 1、Atg5蛋白表达和LC3BⅡ/LC3BⅠ比值显著升高,p62蛋白表达显著下降(P<0.01);与模型组比较,肾康灵组小鼠肾质量增加(P<0.05),血尿素氮和血肌酐含量明显减少(P<0.05,P<0.01),肾组织病理变化减轻,Beclin 1、Atg5蛋白表达和LC3BⅡ/LC3BⅠ明显降低,p62蛋白表达显著升高(P<0.05,P<0.01)。结论肾康灵可能通过调控自噬改善CKD肾损伤,从而延缓CKD进展。
Objective To observe the effects of Shenkangling on renal function and renal autophagy in mice with chronic kidney disease(CKD);To explore the protective mechanism of Shenkangling on renal injury of CKD.Methods Totally 24 C56BL/6 mice were randomly divided into normal group,model group and Shenkangling group,with 8 mice in each group.The CKD mouse model was induced by adenine.After 28 days of intragastric treatment with Shenkangling(1.5 g/kg),the blood urea nitrogen(BUN)and serum creatinine(SCr)were detected;the pathological morphology of kidney tissues was observed by HE staining,Masson staining and glycogen PAS staining,respectively;the changes of renal ultrastructure and autophagosome were observed by transmission electron microscope;the expression of autophagy-related proteins p62,Beclin 1,Atg5 and LC3B in the renal tissue of mice were detected by Western blot.Results Compared with the normal group,the kidneys of the mice in the model group were atrophy,the mass of the kidney decreased significantly(P<0.01),the contents of BUN and SCr significantly increased(P<0.01),the fibrosis lesion in renal tissue was obvious,the proteins expression of Beclin 1,Atg5 and the ratio of LC3BⅡ/LC3BⅠsignificantly increased,and the expression of p62 significantly decreased(P<0.01).Compared with the model group,the mass of the kidney increased significantly(P<0.01),the conents of BUN and SCr of the mice in the Shenkangling group decreased significantly(P<0.05,P<0.01),the pathological changes of kidney tissues were alleviated,the proteins expression of Beclin 1,Atg5 and the ratio of LC3BⅡ/LC3BⅠdecreased,and the expression of p62 significantly increased(P<0.05,P<0.01).Conclusion Shenkangling may improve renal injury of CKD by regulating autophagy,in order to ultimately delay the progression of CKD.
