推拿手法对缺血再灌注大鼠脑神经及自噬相关蛋白Beclin 1表达的影响 被引量:1

摘要 目的观察推拿法对脑缺血再灌注损伤大鼠神经功能及自噬相关蛋白Beclin 1表达的影响。方法 Sprague Dawley大鼠(SD大鼠)共30只,随机分为假手术组、模型组、推拿组,每组分别为10只。模型制备成功后推拿组给予推拿刺激,其余2组不予处理。相应治疗2周,观察记录相应时间点的ZeaLonga神经功能评分。2周后取大鼠脑组织,TTC观察脑梗死面积,TUNEL法检测脑神经细胞凋亡程度,western blot、qPCR检测脑皮质Beclin 1蛋白及mRNA的表达情况。结果 ZeaLonga评分显示,与假手术组比较,模型组的评分明显增高,与模型组比较,推拿组的评分明显降低;TTC染色显示,模型组的脑梗死面积明显高于假手术组与推拿组;TUNEL法显示,推拿组与假手术组的脑神经细胞凋亡指数明显低于模型组。推拿组的Beclin 1蛋白的表达含量较模型组增加;推拿组的Beclin 1 mRNA表达显著高于模型组。结论推拿法可能通过促进Beclin 1的表达来调节自噬和细胞凋亡,改善大鼠神经功能缺损,从而起到对脑缺血再灌注大鼠的保护作用。 Objective To observe the effect of massage on the neurological function and the expression of autophagy-related protein Beclin 1 in rats with cerebral ischemia-reperfusion injury.Methods A total of 30 Sprague Dawley rats (SD rats) were randomly divided into a sham operation group,a model group and a massage group,with 10 rats in each group.After the model was successfully prepared,the massage group was given a massage stimulation,while the other two groups were not treated.After 2 weeks of the corresponding treatment,the ZeaLonga neurological function scores at the corresponding time points were observed and recorded.Two weeks later,the brain tissue of rats was taken and the cerebral infarction area were observed by TTC,and the apoptosis of brain neurons was detected by the TUNEL method,while the expression of Beclin 1 protein and mRNA in the cerebral cortex was detected by Western blot and qPCR.Results The ZeaLonga scores showed that the score of the model group was significantly higher than that of the sham operation group,while the score of the massage group was significantly lower than that of the model group.The TTC staining observation showed that the cerebral infarction area of the model group was significantly higher than that of the sham operation group and the massage group.The TUNEL method showed that the neuronal apoptosis index of the massage group and the sham operation group was significantly lower than that of the model group.The expression of Beclin 1 protein in the massage group increased compared with that in the model group and the expression of Beclin 1 mRNA in the massage group was significantly higher than that in the model group.Conclusion Massage may promote the expression of Beclin 1 to regulate autophagy and apoptosis,improve the neurological deficit,and thus play a protective role in rats with cerebral ischemia reperfusion.
