“秩边透水道”针法对子宫内膜异位症大鼠Beclin-1、LC3调节作用的研究 被引量:5

Study of the Regulatory Effect of Needling Zhibian(BL54)Toward Shuidao(ST28)on Beclin-1 and LC3 in Rats with Endometriosis

摘要 目的观察“秩边透水道”针法对子宫内膜异位症(EMS)大鼠Bclin-1、LC3表达的影响,探讨此针法治疗EMS的可能机制。方法50只雌性SD大鼠随机分为模型组、假手术组、针刺组、激动剂组、针刺加抑制剂组,每组10只。假手术组不造模,不干预,其余各组造模成功后,分别处理。针刺组大鼠予以“秩边透水道”针刺干预;激动剂组予以雷帕霉素(rapamycin)腹腔注射;针刺加抑制剂组在针刺组干预基础上,同时予以3-甲基腺嘌呤(3-methyladenine,3-MA)腹腔注射;模型组和假手术组动物常规饲养。各组均干预3周。取各组大鼠子宫在位、异位内膜组织,苏木素-伊红(HE)染色观察病理学变化;透射电镜观察自噬小体变化;RT-qPCR、Western blot法检测Beclin-1、LC3 mRNA和蛋白的表达。结果HE染色结果显示大鼠子宫内膜上皮都出现萎缩,并伴随纤维组织的增生和毛细血管的减少,其中针刺组和激动剂组变化最为明显。透射电镜结果显示模型组自噬泡减少;针刺组自噬泡较多;激动剂组大鼠子宫内膜细胞中自噬泡较多;针刺加抑制剂组有少量的自噬泡。无论在各组大鼠子宫在位还是异位内膜中,模型组LC3、Beclin-1 mRNA及蛋白表达均明显低于假手术组(P<0.05);各干预组LC3、Beclin-1 mRNA及蛋白表达均明显高于模型组(P<0.05);针刺组、激动剂组LC3、Beclin-1 mRNA及蛋白表达均明显高于针刺加抑制剂组(P<0.05)。结论“秩边透水道”针法可上调大鼠子宫内膜组织内Beclin-1、LC3的表达,改善大鼠子宫内膜细胞防护效应机制。 Objective To observe the effect of needling Zhibian(BL54)toward Shuidao(ST28)on the expression levels of Beclin-1 and LC3 in rats with endometriosis(EMS)and discuss the plausible mechanism of this acupuncture method for EMS.Method Fifty female Sprague-Dawley(SD)rats were randomized into a model group,a sham operation group,an acupuncture group,an agonist group,and an acupuncture+inhibitor group,with 10 rats in each group.The sham operation group received neither the modeling procedure nor the intervention,while the other groups received the corresponding intervention after modeling.Rats in the acupuncture group were given acupuncture method of needling Zhibian toward Shuidao;the agonist group received intraperitoneal injection of Rapamycin;the acupuncture+inhibitor group received intraperitoneal injection of 3-methyladenine(3-MA)and the same intervention in the acupuncture group;the model and sham operation groups received the conventional feeding.The intervention lasted for three weeks in each group.The eutopic and ectopic endometrial tissue was collected from each group of rats.The pathological changes were observed using hematoxylin-eosin(HE)staining;transmission electron microscope was used to observe the change of autophagosomes;RT-qPCR and Western blot were adopted to detect the expression levels of Beclin-1 and LC3 mRNAs and proteins.Result HE staining showed atrophic endometrial epithelia accompanied by the proliferation of fibrous tissue and reduction of capillaries in each group,particularly in the acupuncture and agonist groups.Transmission electron microscope revealed the reduction of autophagic vacuole in the model group;comparatively more autophagic vacuole in the acupuncture group;comparatively more autophagic vacuole in rat’s endometrial cells in the agonist group;and a small amount of autophagic vacuole in the acupuncture+inhibitor group.In both eutopic and ectopic endometria,the expression levels of LC3 and Beclin-1 mRNAs and proteins were significantly lower in the model group than in the sha
