摘要
目的研究马蹄皮多酚的抗氧化活性,并探讨其对人肝癌HepG-2细胞增殖的影响。方法利用超声波提取法制备马蹄皮多酚;采用硫代巴比妥酸法、邻苯三酚氧化法研究马蹄皮多酚的抗氧化活性;MTT法测定其对HepG-2细胞增殖的抑制作用。结果马蹄皮多酚可抑制大鼠红细胞溶血,影响大鼠脑组织脂质过氧化,降低肝组织中丙二醛的含量,并对大鼠超氧阴离子自由基、羟基自由基有极强的清除作用。0.35~1.15 mg·mL^(-1)马蹄皮多酚可抑制HepG-2细胞的增殖,且不影响人胚肺成纤维细胞HFL-1的增殖;浓度为1.15、1.55 mg·mL^(-1)时,HepG-2细胞失去完整结构。结论马蹄皮多酚具有抗氧化活性和抑制HepG-2细胞增殖的作用。
OBJECTIVE To investigate antioxidant activity of polyphenols from water chestnut peel(PWCP),and to explore its effect on proliferation of HepG-2 cells.METHODS The antioxidant activity of PWCP was measured by Thiobarbituric acid method and Pyrogallol oxidation method.The antiproliferative activity on HepG-2 cells was measured by MTT method.RESULTS PWCP could inhibit erythrocyte hemolysis, suppress lipid peroxidation in rat brain tissue, decrease the level of MDA in liver tissue, and scavenge superoxide anion radical and hydroxyl radical.PWCP could inhibit the proliferation of HepG-2 cells at concentration range of 0.35-1.15 mg·mL;,but there was no effect on the proliferation of human embryonic lung fibroblasts HFL-1 normal cells.HepG-2 cells lost their complete structure at the concentration of 1.15 mg·mL;and 1.55 mg·mL;.CONCLUSION PWCP has antioxidant activity and anti-proliferation effect on HepG-2 cells.
