摘要
目的探讨过表达微小RNA(miRNA,miR)-4731-5p对结直肠癌HT-29细胞增殖和凋亡的影响及作用机制。方法将miR-4731-5p类似物(miR-4731-5p组)和阴性对照miR-NC(miR-NC组)转染至结直肠癌HT-29细胞。实时定量聚合酶链反应(qRT-PCR)法检测HT-29细胞miR-4731-5p的相对表达量。采用集落形成实验和流式细胞术分别检测HT-29细胞的增殖水平和凋亡情况。生物信息学软件miRTarBase预测miR-4731-5p的靶基因。qRT-PCR和Western blot检测HT-29细胞靶基因的相对表达量。结果miR-NC组和miR-4731-5p组HT-29细胞中miR-4731-5p的表达分别为(1.05±0.16)和(7.91±1.26),差异有统计学意义(P<0.01)。与miR-NC组相比,miR-4731-5p组HT-29细胞增殖能力明显减弱(P<0.01),细胞凋亡明显增强(P<0.01)。miR-4731-5p的靶基因可能是富含脯氨酸蛋白11(proline-rich protein 11,PRR11)基因。与miR-NC组相比,miR-4731-5p组HT-29细胞中PRR11基因表达明显降低[(0.28±0.11)比(1.02±0.12)](P<0.01)。结论过表达miR-4731-5p可抑制结直肠癌HT-29细胞的增殖、促进HT-29细胞的凋亡,其机制可能是通过抑制PRR11基因表达实现。
Objective To explore the effect of miRNA(miR)-4731-5p overexpression on the proliferation and apoptosis of colorectal cancer HT-29 cells and its mechanism.Methods The miR-4731-5p analog(miR-4731-5p group)and the negative control miR-NC(miR-NC group)were transfected into colorectal cancer HT-29 cells.Real-time quantitative polymerase chain reaction(qRT-PCR)method was used to detect the relative expression of miR-4731-5p in HT-29 cells.Colony formation experiment and flow cytometry were used to detect the proliferation level and apoptosis of HT-29 cells.The bioinformatic software miRTarBase was used to predict the target gene of miR-4731-5p.qRT-PCR and Western blot were used to detect the relative expression of target gene in HT-29 cells.Results The expressions of miR-4731-5p in HT-29 cells of the miR-NC group and miR-4731-5p group were(1.05±0.16)and(7.91±1.26),respectively,with a statistically significant difference(P<0.01).Compared with those in the miR-NC group,the proliferation ability of HT-29 cells in the miR-4731-5p group was significantly weakened(P<0.01),and the cell apoptosis was significantly enhanced(P<0.01).The target gene of miR-4731-5p may be proline-rich protein 11(PRR11)gene.Compared with that in the miR-NC group,the expression of PRR11 gene in HT-29 cells in the miR-4731-5p group was significantly reduced[(0.28±0.11)vs.(1.02±0.12)](P<0.01).Conclusions Overexpression of miR-4731-5p can inhibit the proliferation of colorectal cancer HT-29 cells and promote the apoptosis of HT-29 cells.The mechanism may be achieved by inhibiting the expression of PRR11 gene.
