摘要
采用硅烷化衍生-气相色谱-质谱联用法测定动物源食品中游离氨基酸含量。新鲜猪肉、鸡蛋和奶酪样品用蒸馏水萃取,萃取液经冷冻干燥去除水分后,加入N,O-双(三甲基硅烷基)三氟乙酰胺(BSTFA∶TMCS,99∶1)硅烷化衍生试剂,在100℃条件下硅烷化衍生反应30 min,进行GC/MS分析。结果表明,17种氨基酸的定量标准曲线线性关系良好,R2为0.9884~0.9965。该测定方法前处理简便快速、灵敏度高,能够用于动物源食品中游离氨基酸的检测。
The content of free amino acids in animal-derived foods was determined by silanization derivatization-gas chromatography-mass spectrometry.Fresh pork,egg and cheese samples were extracted with distilled water,and the extract was freeze-dried and added with N,O-bis(trimethylsilyl)trifluoroacetamide(BSTFA∶TMCS,99∶1)at 100℃.After 30 min of the silylation derivatization reaction,GC/MS analysis was performed in the selected ion monitoring(SIM)mode.The results showed that the quantitative standard curve of 17 amino acids had good linearity,with R2 ranging from 0.9884 to 0.9965.This method was quick and simple in pre-processing operation,high in detection sensitivity,and was suitable for the quantitative analysis of free amino acid content in animal-derived foods.
