气相色谱-质谱法分析乳腺癌大鼠尿液中的代谢物 被引量:2

摘要 基于气相色谱-质谱法,建立了乳腺癌大鼠尿液中代谢物的分析方法。构建了乳腺癌大鼠模型。分别收集乳腺癌组、未成瘤组和健康对照组大鼠尿液样本,经脲酶处理后,用甲醇∶乙腈=2∶1(V/V)进行提取,硅烷化试剂N,O-双甲氧基三氟乙酰胺+1%三甲基氯硅烷(BSTFA+1%TMCS)衍生,然后采用气相色谱-质谱法分析尿液样本的代谢谱,并用随机森林(RF)算法对实验结果进行分析。结果表明,三组实验数据被成功分类,并发现甲基丙二酸、3-羟基丁酸、苏糖酸、对羟基苯乙酸、D-麦芽糖等组分在乳腺癌组和健康对照组中具有显著差异。 Based on gas chromatography-mass spectrometry,a urine metabolomics method was developed and applied to rat breast cancer.A breast cancer rat model was constructed,then the urine from rats of the breast cancer group,non-tumor group and healthy group were collected.After being treated with urease,the urine samples were extracted with methanol∶acetonitrile=2∶1(V/V),and then derived with N,O-bis(trimethylsilyl)trifluoroacetamide and trimethylchlorosilane(BSTFA+TMCS,99∶1,V/V).Finally,the urine samples were analyzed by gas chromatography-mass spectrometry,and the metabolic spectra were collected and analyzed by the random forest(RF)algorithm.The results showed that the three sets of experimental data were successfully classified.Especially,methylmalonic acid,3-hydroxybutyric acid,threonic acid,p-hydroxyphenylacetic acid,and D-maltose showed significant differentce between breast cancer group and healthy control group in this experiment.
