摘要
目的:基于磷脂酰肌醇3-激酶(phosphatidylinositol 3-kinase,PI3K)/蛋白激酶B(protein kinase B,Akt)/哺乳动物雷帕霉素靶蛋白(mammalian target of rapamycin,mTOR)信号通路,探讨双参活血颗粒对急性心肌梗死大鼠自噬的调控作用。方法:60只SD大鼠按随机数字表法随机分为正常组、模型组、中药高、中、低剂量组与西药组,采用冠状动脉左前降支结扎术制备心肌梗死大鼠模型,西药组、中药组分别给予盐酸曲美他嗪片、中药双参活血颗粒灌胃连续2周,Western Blot法检测各组大鼠心肌组织中PI3K、Akt、mTOR蛋白表达水平及磷酸化磷脂酰肌醇3-激酶(p-PI3K)、磷酸化蛋白激酶B(p-Akt)、磷酸化哺乳动物雷帕霉素靶蛋白(p-mTOR)的蛋白表达水平。结果:与正常组比较,模型组p-PI3K、p-Akt、p-mTOR蛋白表达水平显著下调(P<0.05),PI3K、Akt、mToR蛋白表达差异无统计学意义(P>0.05);与模型组比较,中药组、西药组p-PI3K、p-Akt、p-mTOR蛋白表达水平均明显上调(P<0.05),其中以西药组上调极为显著,且中药组的上调幅度随给药剂量的增加而增加,PI3K、Akt、mTOR蛋白表达差异无统计学意义(P>0.05)。结论:双参活血颗粒可能通过激活PI3K/Akt/mTOR信号通路抑制过度自噬,从而减轻大鼠心肌梗死晚期损伤。
Objective:To investigate the regulating effect of the Shuangshen Huoxue Granules on the autophagy of acute myocardial infarction(AMI)rats based on the PI3K/Akt/mTOR signaling pathway.Methods:Sixty healthy SD rats were randomly divided into six groups:normal group,model group,Shuangshen Huoxue Granules high dose group,middle dose group,low dose group and western medicine group.The rat models of AMI were established by the ligation of anterior descending branch of left coronary artery.The western medicine group was intragastrically given Trimetazidine hydrochloride tablets and traditional Chinese medicine group was given Shuangshen Huoxue Granules for 2 weeks.The levels of PI3K,p-PI3K,Akt,p-Akt,mTOR and p-mTOR in myocardial tissues of each group were detected by Western Blot.Results:Compared with the normal group,p-PI3K,p-Akt and p-mTOR expression were significantly decreased in the model group(P<0.05),there were no significant differences in the expressions of PI3K,Akt and mTOR(P>0.05);Compared with the model group,p-PI3K,p-Akt and p-mTOR expression were increased in the traditional Chinese medicine group and western medicine group(P<0.05),the western medicine group had the most significant increase.There were no significant differences in the expressions of mTOR,PI3K and Akt(P>0.05).Conclusion:Shuangshen Huoxue Granules may inhibit excessive autophagy by activating PI3K/Akt/mTOR signaling pathway to alleviate late myocardial infarction injury in rats.
