摘要
采用新型荧光衍生试剂2-[2-(7H-二苯并[a,g]咔唑)-乙氧基]-乙基氯甲酸酯(DBCEC-Cl)作为柱前衍生试剂,在非水毛细管电泳模式下实现了15种氨基酸衍生物的基线分离。实验采用毛细管(总长58.5 cm,有效长度50 cm,内径50μm),甲酰胺为溶剂,乙酸铵-乙酸为缓冲体系,290 nm二极管阵列进行检测。探讨了乙酸铵、乙酸的浓度,电压,柱温对氨基酸衍生物分离的影响。结果表明:在乙酸铵浓度70 mmol/L,乙酸浓度0.65 mol/L,分离电压30 kV,柱温18℃,进样8 s的最优条件下,氨基酸衍生物得到了基线分离。
A simple and rapid method was developed for the separation of 15 amino acids derivatives using 2-[2-(7H-dibenzo[a,g]carbazole-ethoxy)]-ethyl chloroformate(DBCEC-Cl)as derivatization reagent by nonaqueous capillary electrophoresis.A 58.5 cm×50μm i.d.(50 cm effective length)untreated fused-silica capillary was used.The detection was performed with diode array at 290 nm.Formamide was used as the background electrolyte solvent while ammonium acetate-acetic acid as the buffer.Effects of some factors were investigated.The optimum separation conditions were as follows:with 70 mmol/L ammonium acetate and 0.65 mol/L acetic acid,voltage was 30 kV,column temperature was at 18℃.The samples were atmospherically introduced with an injection time of 8 s.The results indicated that derivatized amino acids achieved baseline resolution within 30 min under the proposed conditions.
