摘要
目的利用分子生物学技术,通过体内实验,观察和胃反流康对实验性胆汁反流性胃炎模型大鼠胃黏膜p38MAPK蛋白及胃黏膜细胞COX-2mRNA表达的影响。方法将60只SPF级大鼠随机分为6组,即空白组、模型组、和胃反流康高、中、低剂量组(54.6 g·kg^(-1),27.3 g·kg^(-1),13.7 g·kg^(-1))、阳性药物(吗丁啉)组,每组10只。配制反流液,除空白组外,其余各组均采用灌胃方式建立胆汁反流性胃炎大鼠模型。分别通过Western blot法及实时荧光定量PCR法,检测大鼠胃窦部胃黏膜组织中p38MAPK蛋白及胃黏膜细胞COX-2mRNA的表达,观察和胃反流康对胆汁反流性胃炎大鼠胃黏膜p38MAPK蛋白及胃黏膜细胞COX-2mRNA表达水平的影响。结果和胃反流康可显著抑制大鼠胃黏膜p38MAPK蛋白及胃黏膜细胞COX-2mRNA的表达水平(P<0.01)。结论和胃反流康可显著抑制胆汁反流性胃炎大鼠胃黏膜p38MAPK蛋白及胃黏膜细胞COX-2mRNA的表达,减轻反流液对胃黏膜的炎性损害,具有防止BRG模型大鼠胃黏膜肠上皮化生及不典型增生等癌前病变发生的作用。
Objective To observe the effects of Hewei Fanliu Kang on the expression of the p38MAPK protein and COX-2mRNA in gastric mucosal cells of experimental bile reflux gastritis rats in vivo by molecular biology techniques.Methods A total of sixty SPF rats were randomly divided into 6 groups,namely the blank group,the model group,the Hewei Fanliu Kang high-,medium-and low-dose groups(54.6 g·kg^(-1),27.3 g·kg^(-1),13.7 g·kg^(-1)),and the positive drug group(morpholine),with 10 rats in each group.The bile reflux gastritis rat model was established by gavage of the prepared reflux liquid in all the groups except the blank group.Western blot and real-time fluorescent quantitative PCR were used to detect the expression of the p38MAPK protein and COX-2mRNA in gastric mucosal tissues of rat gastric antrum and to observe the effect on their expression levels.Results Hewei Fanliu Kang significantly inhibited the expression levels of the p38MAPK protein and COX-2mRNA in gastric mucosal cells of rats(P<0.01).Conclusion Hewei Fanliu Kang can significantly inhibit the expression of the p38MAPK protein and COX-2mRNA in gastric mucosa of bile reflux gastritis rats,reduce the inflammatory damage to gastric mucosa caused by reflux fluid,and prevent the occurrence of intestinal metaplasia,atypical hyperplasia and other precancerous lesions in gastric mucosa of BRG model rats.
