多发性硬化患者唾液微生物菌群多样性的研究

Study of Salivary Microflora in Patients with Multiple Sclerosis

摘要 目的分析多发性硬化(MS)患者唾液微生物菌群,探讨MS患者口腔微生物菌群构成特点与MS发病的相关性。方法选取2015年1月至2018年12月就诊的非急性加重期MS患者30例为MS组,另选择30例健康者为对照组。收集研究对象非刺激性唾液,提取唾液DNA后进行高通量测序并行数据分析。结果MS组唾液微生物α多样性中菌群丰度指数ACE高于对照组(P<0.05)。共检测到13个菌门,其中5个主要优势菌门依次为:厚壁菌门、变形菌门、拟杆菌门、梭杆菌门、放线菌门。MS组唾液中拟杆菌门低于对照组,而梭杆菌门和螺旋体门含量高于对照组(均P<0.05)。共检测到161个菌属,优势菌属12个:MS组链球菌属、卟啉单胞菌属、密螺旋体属、月形单胞菌属、产线菌属、石胡荽属及小类杆菌属含量均高于对照组(均P<0.05)。而MS组普雷沃菌属、扁平杆菌属及莫拉菌属含量低于对照组(均P<0.05)。MS组微生物群落近似度高于对照组。结论与健康人群比较,MS患者口腔唾液具有独特微生物多样性结构,其口腔微生物菌群结构有明显区别。 Aim To discuss the relationship between the characteristics of oral microflora and multiple sclerosis(MS)by analyzing the salivary microflora of MS patients.Methods From January 2015 to December 2018,30 non-acute exacerbation MS patients in the Third Affiliated Hospital of Qiqihar Medical University were recruited and 30 healthy people were selected as a control group.Non irritant saliva was collected,and saliva DNA was extracted for high-throughput sequencing and data analysis.Results The ace of salivary microbialαdiversity in MS group was higher than that in the control group,and the difference was statistically significant(P<0.05).Among thirteen phyla detected,five main dominant phyla were as follows:Firmicutes,Proteobacteria,Bacteroides,Fusobacteria,Actinobacteria(P<0.05).The content of Fusobacteria and Spirochaetes in saliva of MS group were higher than those of the control group,but the Bacteroides is lower than those of the control group(P<0.05).Among 161 genera detected,there were twelve dominant genera.In MS group,the content of Streptococcus,Porphyromonas,Treponema,Selenomonas,Filifactor,Centipeda and Dialister were higher than those in the control group(P<0.05).The content of Prevotella,Planobacterium and Moraxella in MS group was lower than that in the control group,and the difference was statistically significant(P<0.05).The similarity of microbial community in MS group was higher than that in the control group.Conclusion Compared with the healthy population,the oral saliva of MS patients has a unique microbial diversity structure.
