pH微环境对成牙本质细胞增殖行为的影响

摘要 目的探讨pH微环境对成牙本质细胞增殖活性的影响。方法以永生化成牙本质细胞系小鼠牙乳头细胞-23(murine dental papilla cells-23,MDPC-23)为研究对象,利用盐酸、氢氧化钠溶液调节培养基pH,模拟酸、碱及生理环境的不同pH,并检测不同pH培养基中MDPC-23细胞的增殖活性,以分析pH微环境对其生长、增殖行为的影响。结果成牙本质细胞MDPC-23在不同pH的微环境下呈现出不同的形态特征,其细胞形态随着培养基pH值的增高呈现短梭形、聚集生长的趋势;其增殖活性在酸性环境下得到抑制,并随着pH的增大而逐渐增加。结论龋坏部位由于细菌产酸代谢产生的低pH微环境可引起成牙本质细胞生理活动改变,pH微环境能够调控成牙本质细胞形态和增殖活性。 Objective To clarify the effects of pH microenvironment on proliferative activity of odontoblast cells.Methods The murine dental papilla cells-23(MDPC-23)were taken as study subjects.Hydrochloric acid and sodium hydroxide were used to adjust the pH of culture medium to simulate the pH value of acidic,alkaline and physiological environments respectively.The proliferative activity of MDPC-23 cells under different pH conditions was determined,and the interrelation between them was analyzed.Results MDPC-23 cells showed different morphological characteristics under different pH microenvironments.The cell morphology showed a trend to be short spindle shape and to grow accumulationally with the increase of pH value of medium.The proliferative activity of MDPC-23 cells was inhibited under an acidic environment,and gradually increased with the increase of pH value.Conclusion Low pH in the carious cavity,caused by the acid-producing metabolism of bacteria,can regulate the morphology and the proliferative activity of odontoblast cells.
