摘要
目的建立了一种快速测定黄芪桂枝五物汤和桂枝水提物中桂皮酸的UFLC方法。方法实验中使用C18色谱柱,以0.1%磷酸水(A)-乙腈(B)为流动相进行梯度洗脱,流速为0.4 mL/min,柱温为35℃,检测波长为290 nm,分析时间仅为15 min。结果桂皮酸的线性范围为0.5~20 mg/L(r=0.9997),所建立的UFLC测定方法具有良好的线性、精密度、稳定性、重复性和回收率。桂皮酸在黄芪桂枝五物汤和桂枝水提物中的含量分别为0.138~0.157 g/L和0.179~0.195 g/L。结论桂皮酸在黄芪桂枝五物汤中比在桂枝水提物中的含量少,其他药材成分对桂皮酸的提取具有抑制作用。
Objective To establish a UFLC method for the rapid determination of cinnamic acid in Huangqi Guizhi Wuwu Decoction and Ramulus Cinnamomi aqueous extract.Methods In the experiment,C18 chromatographic column was used for gradient elution with 0.1%phosphoric acid water(a)-acetonitrile(b)as mobile phase.The flow rate was 0.4 mL/min,the column temperature was 35℃,the detection wavelength was 290 nm,and the analysis time was only 15 min.Results The linear range of cinnamic acid was 0.5-20 mg/L(r=0.9997).The established UFLC method had good linearity,precision,stability,repeatability and recovery.The contents of cinnamic acid in Huangqi Guizhi Wuwu Decoction and Ramulus Cinnamomi aqueous extract were 0.138-0.157 g/L and 0.179-0.195 g/L respectively.Conclusion The content of cinnamic acid in Huangqi Guizhi Wuwu Decoction is less than that in Ramulus Cinnamomi aqueous extract.Other medicinal materials have inhibitory effects on the extraction of cinnamic acid.
