大黄素对免疫应激仔猪回肠黏膜形态、抗氧化能力、紧密连接蛋白及炎性因子mRNA表达的影响 被引量:2

Effects of Emodin on Morphology,Antioxidant Capacity,Tight Junction Protein and Inflammatory Factor mRNA in Ileum Mucosa of Immune-stressed Piglets

摘要 试验旨在研究大黄素(emodin,ED)对脂多糖(lipopolysaccharide,LPS)诱导的免疫应激断奶仔猪肠道的黏膜形态、抗氧化能力、紧密连接蛋白及炎性因子mRNA的影响。试验挑选健康状况良好、胎次相近的28日龄左右的三元杂交(杜×长×大)断奶仔猪24头,采用2×2双因素随机试验设计,分为日粮(基础日粮或日粮中添加300 mg/kg大黄素)和应激(生理盐水对照或脂多糖注射)两个处理,4个组设计如下:①CON组,饲喂基础日粮;②ED组,日粮中添加300 mg/kg大黄素;③LPS应激组,注射LPS、饲喂基础饲粮;④LPS+ED组,注射LPS、饲喂基础饲粮并添加300 mg/kg大黄素。在试验第21 d LPS应激组和LPS+大黄素组腹膜注射100μg/kg·BW LPS,建立免疫应激模型,对照组和大黄素组注射同等剂量的灭菌生理盐水。试验期为21 d。结果表明:对比基础日粮组,饲粮中添加300 mg/kg ED仔猪的绒毛高度和V/C指标显著升高(P<0.05);在饲粮中添加ED后,则会影响过氧化氢酶(CAT)活力的升高、总抗氧化能力(T-AOC)的上升(P<0.05);饲粮中添加ED显著提高了CAT的mRNA相对表达(P<0.05),ED的添加使得IL-6和TNF-α的mRNA相对表达量和含量显著降低,饲粮中添加ED会显著提高肠黏膜闭锁小带蛋白(ZO-1)、闭锁蛋白(Occludin)的相对表达量(P<0.05)。综上所述,饲粮中添加ED可以改善回肠黏膜形态,提高回肠紧密连接蛋白表达,提高回肠抗氧化性能和抗炎性能,保护肠道的健康。 This experiment was conducted to investigate the effects of emodin(ED)on intestinal mucosal morphology,antioxidant capacity,tight junctive protein and inflammatory factor mRNA in weaned piglets under immune stress induced by lipopolysaccharide(LPS).In the experiment,24 three-way cross(Du×Long×Large)weaned piglets with good health and similar parity were selected.The 2×2 two-factor randomized trial design was adopted and divided into diet(basic day).There were two treatments(300 mg/kg emodin added to food or diet)and stress(normal saline control or lipopolysaccha⁃ride injection).The four groups were designed as the followsing:①CON group,fed with basal di⁃et;②ED group,300 mg/kg emodin was added to the diet;③LPS stress group,LPS injection,and bas⁃al diet;④LPS+ED group,LPS injection,basal diet and 300 mg/kg rhubarb white.On the 21st day,100μg/kg·BW LPS was injected intraperitoneally in the LPS stress group and the LPS+emodin group.The control group and the emodin group were injected with the same dose of sterilized normal saline.The test period was 21 days.In conclusion,dietary supplemented with ED could improve the morphology of ileum mucosa,increase the expression of ileum tight junction protein,improve the ileum antioxidant capacity and anti-inflammatory activity,and protect the health of the intestine.
