菊苣中菊粉和低聚果糖调节脂质代谢的生信分析 被引量:4

Bioinformatics analysis of regulation of lipid metabolism by inulin and fructooligosaccharide in Cichorium intybus L.

摘要 目的:通过生物信息学技术研究菊苣中菊粉与低聚果糖在小鼠结肠中调节脂质代谢的共同作用途径,探讨菊苣降脂作用的机制。方法:以菊粉和低聚果糖影响小鼠结肠的研究为例,采用生物信息技术,对GEO数据库中含有菊粉和低聚果糖的基因芯片GSE59494进行筛选分析,对小鼠结肠上皮细胞共同差异基因进行GO功能注释和KEGG通路分析,分析二者调节脂代谢的共有机制。结果:菊粉和低聚果糖通过上调Adipoq等脂代谢基因表达和激活PPARs、AMPK相关信号通路调节脂代谢,为菊苣防治肥胖、高脂血症等代谢性疾病提供参考。结论:菊苣降脂作用机制可能与上调Adipoq等脂代谢基因表达及激活PPARs等脂代谢信号通路有关。 Objective:To study the common way of regulating lipid metabolism of inulin and fructooligosaccharide,components of Cichorium intybus L.,in the mouse colon through bioinformatics technology,and to explore the mechanism of the lipid-lowering effect of Cichorium intybus L..Methods:In this study,inulin and fructooligosaccharide were used to study the effects of inulin and fructooligosaccharide on the colon of mice.Using bioinformation technology,GSE59494 gene chip containing inulin and fructooligosaccharide in GEO database was screened and analyzed.GO functional annotation and KEGG pathway analysis were performed on the common differentially genes of mouse colonic epithelial cells to analyze the common mechanism of inulin and fructooligosaccharide in regulating lipid metabolism.Results:Inulin and fructooligosaccharide play a lipid-lowering role in the colon of mice by up-regulating the expression of Adipoq and other lipid metabolism genes and activating the PPARs and AMPK signaling pathways,providing reference for the prevention and treatment of obesity,hyperlipidemia and other metabolic diseases with chicory.Conclusion:The mechanism of Cichorium intybus L.may be related to the up-regulation of Adipoq and other lipid metabolism genes and the activation of PPARs and other lipid metabolism signaling pathways.
