摘要
目的:研究右美托咪定对使用七氟醚进行麻醉的大鼠下丘脑内磷酸化Ca^(2+)/CaM-依赖性蛋白激酶Ⅱα(pCaMKⅡα)表达的影响。方法:选择经侧脑室置管的大鼠20只作为研究对象。随机将其分为对照(control,Con)组和右美托咪定(dexmedetomidine,Dex)组。经Con组大鼠的侧脑室小管注入无菌生理盐水0.1 mL,经Dex组大鼠的侧脑室小管注入浓度为20μg/mL的右美托咪定0.1 mL。注药30 min后用七氟醚对两组大鼠进行麻醉,在麻醉30 min时取其下丘脑组织,检测其中pCaMKⅡɑ的表达量,并在同一时间点取其脑电波频谱进行分析。结果:与Con组大鼠相比,Dex组大鼠的抑制性脑电波增多(P<0.05),兴奋性脑电波减少(P<0.05)。用七氟烷麻醉30 min时,与Con组大鼠相比,A组(给予右美托咪定30 min时)大鼠下丘脑内pCaMKⅡɑ蛋白的表达量更低(P<0.05)。与Con组大鼠、A组大鼠相比,B组(给予右美托咪定后用七氟醚麻醉30 min时)大鼠下丘脑内pCaMKⅡɑ蛋白的表达量更低(P<0.05)。结论:右美托咪定可对使用七氟醚进行麻醉大鼠的脑电波产生抑制,还可抑制麻醉过程中其下丘脑内pCaMKⅡɑ的表达。
Objective:To study the effects of dexmedetomidine on the expression of phosphorylated Ca^(2+)/CAM-dependent protein kinaseⅱα(pCaMKⅱα)in hypothalamus of sevoflurane anesthesia rats.Methods:Twenty rats were selected as the study subjects.They were randomly divided into control(Con)group and Dexmedetomidine(Dex)group.Rats in Con group were injected with 0.1 mL sterile normal saline via lateral ventricular tubules,and rats in Dex group were injected with 0.1 mL dexmedetomidine at a concentration of 20μg/mL through lateral ventricular tubules.The rats in both groups were anesthetized with sevoflurane 30 min after injection.The hypothalamus tissue was taken from the rats 30 min after anesthesia to detect the expression level of pCaMKⅱɑ,and the brain wave spectrum was taken from the rats at the same time point for analysis.Results:Compared with Con group,inhibitory eeg was increased(P<0.05)and excitatory EEG was decreased(P<0.05)in Dex group.After 30 min of sevoflurane anesthesia,compared with Con group,the expression of pCaMKⅱɑprotein in hypothalamus of rats in group A(given dexmedetomidine for 30 min)was lower(P<0.05).Compared with Con group and A group,the expression of pCaMKⅱɑprotein in hypothalamus of rats in group B(treated with dexmedetomidine and sevoflurane anesthesia for 30 min)was lower(P<0.05).Conclusion:Dexmedetomidine can inhibit brain wave and pCaMKⅱɑexpression in hypothalamus of sevoflurane anesthe sia rats.
