摘要
目的采用超高效液相色谱-四级杆飞行时间串联质谱(UPLC-Q-TOF-MS)技术分析百事乐胶囊中的化学成分。方法采用Waters ACQUITY UPLC BEH C_(18)(2.1 mm×100 mm,1.7μm)色谱柱,以乙腈-0.1%甲酸水溶液为流动相进行梯度洗脱,流速为0.4 mL·min^(-1),柱温为35℃,进样量为3μL;采用电喷雾离子源,在正、负离子模式下采集数据。运用MassLynx V4.1软件,根据化合物的相对保留时间、准分子离子峰和碎片离子峰等信息,结合对照品和相关文献资料进行化学成分分析。结果在百事乐胶囊中鉴定出了42个化学成分,主要包括有机酸类、黄酮类、皂苷类、萜类和姜黄素类等,并对化合物的药材来源进行了归属。结论建立的方法能够快速、准确、系统地分析百事乐胶囊多种化学成分,较全面地阐述了百事乐胶囊的化学成分信息,为该方的质量控制及药效物质基础研究提供了理论基础。
Objective To study the chemical constituents in Baishile Capsules by ultra-performance liquid chromatography-quadrupole-time of flight-mass spectrometry(UPLC-Q-TOF-MS).Methods The separation was performed on an Waters ACQUITY UPLC BEH C_(18)(2.1 mm×100 mm,1.7μm)by a gradient elution of acetonitrile-0.1%formic acid solution.The flow rate was 0.4 mL·min^(-1)and the column temperature was 35℃,the injection volume was 3μL.Electrospray ionization was applied and the data were collected via positive and negative ion modes.By using MassLynx V4.1 software,the chemical constituents were analyzed based on the retention time,excimer ion peak and fragment ion peak of the compounds,as well as comparison with reference substances and literature data.Results A total of 42 chemical constituents in Baishile Capsules were identified,including organic acids,flavonoids,saponins,terpenoids and curcumin,and the medicinal sources of the compounds were assigned.Conclusion The established method can quickly,accurately and systematically identify various chemical components of Baishile Capsules.The information of chemical constituents in Baishile Capsules is comprehensively expounded.The study establishes a foundation for the research of quality control and material foundation of efficacy of Baishile Capsules.
