复发缓解型多发性硬化患者外周血miR-132、miR-146a与认知障碍的关系 被引量:5

Relationship between peripheral blood miR-146a and miR-132 and cognitive dysfunction in patients with relapse-remission multiple sclerosis

摘要 目的:探究复发缓解型多发性硬化(RRMS)患者外周血清miR-132、miR-146a与认知障碍(CI)的关系。方法:连续入组80例RRMS患者,根据简明精神状态量表(MMSE)评分将其分为认知障碍组(CI组)39例(其中轻度CI组18例,中度CI组13例,重度CI组8例)及无CI组41例。RT-PCR法检测各组血清中miR-132、miR-146a水平,并对相关结果进行比较分析。结果:RRMS患者CI组的外周血清中miR-132、miR-146a水平均高于无CI发生组(P<0.05),而重度CI组患者上述指标水平较其他组升高更明显(P<0.05),相关性分析发现RRMS伴有CI组的miR-132、miR-146a水平与MMSE评分均呈负相关(r 2分别为0.9076、0.9167,P均<0.05),且miR-132、miR-146a是RRMS患者CI发生的独立危险因素(OR=1.356,95%CI:1.038~1.771;OR=5.218,95%CI:2.634~10.340)。结论:RRMS患者外周血miR-132及miR-146a水平与认知功能障碍的严重程度有关,高表达的miR-132及miR-146a是RRMS CI发生的独立危险因素。 Objective:To explore the relationship between peripheral blood miR-132 and miR-146a and cognitive impairment(CI)in patients with relapse-remission multiple sclerosis(RRMS).Methods:A total of 80 patients with RRMS were consecutively enrolled.According to the simple mental state scale(MMSE)scores,39 patients were divided into CI group(18 in mild CI group,13 in moderate CI group and 8 in severe CI group)and 41 in non-CI group.Serum levels of miR-132 and miR-146a in each group were de-tected by RT-PCR,and the relevant results were compared and analyzed.Results:Levels of miR-132 and miR-146a in peripheral blood serum of RRMS patients with CI were higher than those in group without CI(P<0.05),and level of these indicators was more significantly increased in patients with severe CI than in other groups(P<0.05).Correlation analysis showed that the miR-132 and miR-146a levels in RRMS group with CI were negatively correlated with MMSE scores(r 2 were 0.9076 and 0.9167,respectively,P<0.05);in addition,miR-132 and miR-146a were independent risk factors for the occurrence of CI in RRMS patients(OR=1.356,95%CI:1.038~1.771;OR=5.218,95%CI:2.634~10.340).Conclusion:Levels of miR-132 and miR-146a in peripheral blood of RRMS patients are related to the severity of cognitive impairment,and the high expression of miR-132 and miR-146a are independent risk factors for occurrence of RRMS CI.
