摘要
目的观察青石止痒软膏对小鼠特应性皮炎模型皮损部位信号素3A(semaphorin3A,Sema3A)/神经生长因子(nerve growth factor,NGF)及相关分子表达的影响。探讨青石止痒软膏治疗特应性皮炎的作用机制。方法采用2,4-二硝基氯苯诱导小鼠特应性皮炎模型。采用随机数字表法将56只小鼠随机分为空白组、模型组、阴性对照组、阳性对照组、青石止痒低、中、高浓度组,每组8只,连续给药7天后取材。观察各组小鼠特应性皮炎积分(scoring atopic dermatitis,SCORAD);取皮损部位皮肤组织,以苏木精—伊红染色法观察组织病理变化;取皮损部位皮肤组织匀浆,以实时定量荧光聚合酶链式扩增反应定量法检测microRNA-145基因表达;蛋白免疫印迹法检测Sema3A、P21活化激酶(p21-activated kinase,PAK)蛋白表达;酶联免疫吸附法测定白细胞介素31(interleukin-31,IL-31)、NGF、神经生长因子4(neurotrophin-4,NT-4)的表达水平。结果(1)与模型组、阴性对照组相比,治疗后阳性对照组及青石止痒各组SCORAD评分降低(P<0.05),其中青石止痒中浓度组与阳性对照组SCORAD评分无统计学差异(P>0.05);(2)组织病理切片观察发现阳性对照组及青石止痒各浓度组病理学改变较模型组均有改善,与SCORAD评分一致;(3)与空白组相比,模型组、阴性对照组、青石止痒高、低浓度组的microRNA-145表达水平降低(P<0.05),模型组、阴性对照组、青石止痒高、低浓度组的皮肤Sema3A蛋白表达水平降低(P<0.05)、PAK蛋白表达水平升高(P<0.05),模型组、阴性对照组及青石止痒高浓度组皮肤NGF含量升高(P<0.05),模型组NT-4含量升高(P<0.05),模型组、阴性对照组IL-31含量升高(P<0.05);与模型组相比,青石止痒高、中浓度组的microRNA-145表达水平升高(P<0.05),阳性对照组、青石止痒中、低浓度组Sema3A蛋白表达水平升高(P<0.05),PAK蛋白表达水平降低(P<0.05),阳性对照组、青石止痒各浓度组NGF含量�
Objective To observe the effects of Qingshi Zhiyang(QSZY)ointment on Sema3A/NGF and related signaling molecules in atopic dermatitis model mice.To explore the mechanism of treating atopic dermatitis with Qingshi Zhiyang ointment.Methods The atopic dermatitis mice model was induced by DNCB.56 mice were randomly divided into blank group,model group,positive control group,negative control group,QSZY ointment high concentration,medium concentration,low concentration group(n=8 in each group),with the method of random number table,after 7days continuous medicate.The skin tissues were taken for observation.Comparing the SCORAD and results of skin histopathological morphologic with HE staining in different groups;the mRNA expression of microRNA-145 in skin tissue was detected with Real-time PCR;the expression of Sema3A,PAK protein in skin tissues were measured with Western blotting assay;the levels of IL-31,NGF and NT-4 in skin tissues were detected by ELISA.Results(1)Compared with the model group and the negative control group,the SCORAD results in positive control group and each concentration of QSZY groups were lower(P<0.05).There were no statistical difference in SCORAD results between positive control group and QSZY ointment medium concentration group(P>0.05);(2)Compared with the model group,the pathological morphologic changes of skin tissues by HE staining in the positive control group and each concentration group of QSZY were improved.(3)Compared with the blank group,the expression of microRNA-145 was decreased in model group,negative control group and QSZY ointment high,low concentration groups(P<0.05);the expression of Sema3A protein was decreased and PAK protein was increased in the model group,negative control group and QSZY ointment high,low concentration groups(P<0.05);the content of NGF was increased in the model group,negative control group and QSZY ointment high concentration group(P<0.05),the content of NT-4 was increased in the model group(P<0.05),the content of IL-31 was increased in the model group a
