摘要
目的:研究苦参素对胰腺癌增殖和侵袭能力的影响及机制。方法:在体外培养胰腺癌细胞株,不同浓度的苦参素在不同时间点干预胰腺癌细胞,在现象层面,应用MTT法检测胰腺癌细胞的增殖能力;应用Transwell小室检测细胞的侵袭能力。在机制层面,应用Real time PCR和Western blot在转录水平和蛋白水平检测程度,并通过基因转染技术构建高表达sHH的胰腺癌细胞株,进一步验证苦参素对胰腺癌增殖和侵袭能力的影响及sHH通路的作用。结果:高浓度组(40μmol/L)苦参素与低浓度组(20μmol/L)相比,明显抑制胰腺癌细胞的增殖和侵袭能力。在mRNA和蛋白水平,苦参素可明显抑制肿瘤细胞sHH信号通路,降低sHH(P<0.05)和GLI-1的表达水平(P<0.05)。构建高表达sHH的胰腺癌细胞株Panc-1-sh-sHH,苦参素对胰腺癌细胞增殖和侵袭能力的抑制程度明显强于高表达sHH的胰腺癌细胞Panc-1-sh-sHH。结论:苦参素通过抑制sHH信号通路,减弱胰腺癌细胞增殖和侵袭能力。
Objective:To study the effect and mechanism of matrine on the proliferation and invasion in pancreatic cancer cells.Methods:Pancreatic cancer cells were used in vitro,which were intervened by matrine with different concentrations at different time points.MTT assay was carried to examine the effects of matrine on the proliferation ability in pancreatic cancer cells.The effect of matrine on the invasion ability of pancreatic cancer cells was assessed by Transwell chamber.The expression of invasive factors in pancreatic cancer cells was measured by Real-time PCR and Western blot at the mechanism level.Pancreatic cancer cells with high sHH expression were constructed by gene transfection,which further used to verify the effects of matrine and the mechanism of sHH signaling pathway on the proliferation and invasion ability in pancreatic cancer cells.Results:Compared with the low concentration group(20μmol/L),the proliferation and invasion ability of pancreatic cancer cells were significantly inhibited by matrine with the high concentration group(40μmol/L).The protein and mRNA expression of sHH and GLI-1 in pancreatic cancer cells were significantly decreased by matrine with the inhibition of sHH signaling pathway.The pancreatic cancer cell line Panc-1-sh-sHH with high expression of sHH was constructed,the inhibition degree to the proliferation and invasion ability of matrine in pancreatic cancer cells is higher than that of the pancreatic cancer cell line Panc-1-sh-sHH with high expression of sHH.Conclusion:The ability of proliferation and invasion in pancreatic cancer cells was reduced by matrine through the inhibition of sHH signaling pathway.
