摘要 目的:探究自噬在肝癌SMMC-7721细胞DNA放射损伤修复中的影响。方法:用X射线照射肝癌SMMC-7721细胞,细胞分为对照(NC)组、单纯照射(IR)组、IR+自噬激活剂雷帕霉素(Rapamycin)组、IR+自噬抑制剂三甲基腺嘌呤(3-MA)组。彗星实验分析DNA损伤,Western blotting和细胞免疫荧光分别检测LC3B和γ-H2AX蛋白表达。结果:与NC组相比,随着照射剂量增加,IR组DNA损伤严重,γ-H2AX和LC3B蛋白表达升高(P<0.05)。与IR组相比,IR+Rapamycin组γ-H2AX蛋白表达降低,IR+3-MA组γ-H2AX蛋白表达升高(P<0.05)。结论:照射可引起肝癌SMMC-7721细胞DNA损伤,同时可诱导自噬激活;自噬可促进DNA损伤修复。 Objective:To investigate the effect of autophagy in ionizing radiation(IR)-induced DNA damage repair in hepatocellular carcinoma(HCC)SMMC-7721 cells.Methods:Hepatocellular carcinoma cells SMMC-7721 were divided into control group(NC),IR group,IR+Rapamycin(autophagy activator)group and IR+3-MA(autophagy inhibitor)group.Comet assay was used to evaluate the DNA damage.Western blotting and immunofluorescence were used to detect the expressions of LC3B andγ-H2AX.Results:Compared with NC group,DNA damage was serious in IR group,and the expressions ofγ-H2AX and LC3B protein increased with the increase of irradiation dose(P<0.05).Compared with IR group,theγ-H2AX expression was decreased in IR+Rapamycin group,while it was increased in IR+3-MA group(P<0.05).Conclusion:IR can induce DNA damage in HCC SMMC-7721 cells and activate autophagy;autophagy can promote the repair of DNA damage.
