荆芥提取物抗UVA诱导HaCaT角质细胞的光老化作用 被引量:2

Anti-photoaging effects of the Schizonepeta tenuifolia extract on UVA-induced photoaging in HaCaT keratinocytes

摘要 基于UVA辐射HaCaT细胞构建光老化模型,探讨荆芥提取物(STE)的抗光老化作用及机制。福林酚法测定STE总酚含量为(111.09±2.30)mg/g没食子酸当量;DPPH自由基的半数清除率浓度(SC50)为(0.18±0.04)mg/mL,ABTS抗氧化试验结果表明STE抗氧化活性为(3.15±0.06)mmol/g Trolox当量;ELISA试验结果表明STE能够减少HaCaT细胞内反应性氧自由基(ROS)的表达;利用四唑盐比色法(MTT)检测细胞生存率,表明STE能提高UVA辐照后细胞的存活率;蛋白免疫印迹(Western blot)结果表明0.2 mg/mL STE能够显著抑制p-Histone H2AX-S139、p-atm、HSP70、Cleaved caspase-3、p-P38/MAPK、MMP-1和增加Caspase-3、Bcl-2蛋白表达。STE具有抗光老化作用,通过降低UVA诱导HaCaT细胞内ROS的产生,减轻ROS对细胞DNA损伤、抗细胞凋亡、调控热休克蛋白和基质金属蛋白酶信号通路。 The anti-photoaging efficacy of Schizonepeta tenuifolia extract(STE)was evaluated based on the photoaging model which was established by UVA irradiation upon HaCaT.The total phenolic content of STE was(111.09±2.30)mg/g relative to gallic acid as determined by Folin-phenol Reagent.STE exerted significant antioxidant activity,indicative of the half scavenging concentration(SC50)of DPPH(1,1-diphenyl-2-picrylhydrazyl)of(0.18±0.04)mg/mL.The Trolox equivalent antioxidant activity of STE was(3.15±0.06)mmol/g by ABTS(2,2'-azino-bis(3-ethylbenzothiazoline-6-sulfonic acid))assays.In the enzyme linked immunosorbent assay(ELISA),STE could obviously reduce the production of intracellular reactive oxygen species(ROS)in HaCaT cells after UVA irradiation.STE could improve the cell survival rate after UVA irradiation as measured by Tetrazolium salt colorimetry(MTT).The results of Western blot showed that STE could effectively inhibit the expression of p-Histone H2AX-S139,p-atm,HSP70,Cleaved caspase-3,p-P38/MAPK and MMP-1 and increase the expression of Caspase–3 and Bcl-2 proteins compared with the vehicle-treated group.In conclusion,STE exerted obvious anti-photoaging activity by reducing the production of intracellular ROS,alleviating the ROS-induced intracellular DNA damage,antiapoptotic effect,and regulating the signaling pathways of heat shock proteins(HSP)and matrix metalloproteinase(MMP).
