摘要
目的:建立高效液相色谱(HPLC)一测多评法同时对血府逐瘀胶囊中芍药苷、柚皮苷、芸香柚皮苷、橙皮苷、新橙皮苷、甘草苷、甘草酸铵的含量进行定量分析。方法:以Agilent TC-C18(4.6 mm×250 mm,5μm)为色谱柱;柱温为30℃;波长为220 nm;乙腈-0.05%磷酸水溶液为流动相,梯度洗脱;流速为0.8 mL/min。以芍药苷为内参物,建立其他6种指标性成分的相对校正因子(RCFs)及计算各成分的含量。结果:7种指标性成分的质量浓度分别在2.778~55.510μg·mL^(-1)(r=0.9999)、2.333~46.664μg·mL^(-1)(r=0.9998)、1.615~32.290μg·mL^(-1)(r=0.9999)、3.117~62.343μg·mL^(-1)(r=0.9997)、2.225~44.692μg·mL^(-1)(r=0.9998)、3.685~73.701μg·mL^(-1)(r=0.9998)、1.261~25.224μg·mL^(-1)(r=0.9999)范围内线性关系良好,平均回收率分别为99.88%(RSD=1.12%)(n=9)、101.80%(RSD=1.10%)(n=9)、105.07%(RSD=0.53%)(n=9)、98.98%(RSD=1.21%)(n=9)、101.22%(RSD=1.23%)(n=9)、100.99%(RSD=1.31%)(n=9)、98.79%(RSD=1.02%)(n=9),一测多评法计算5批血府逐瘀胶囊中7种成分的含量与外标法实测值间均无明显差异。结论:建立的一测多评法同时测定7种成分的含量,结果准确、可靠,可用于血府逐瘀胶囊的质量控制。
Objective:To establish a quantitative analysis of multi-components by single-marker(QAMS)method for simultaneous determination of paeoniflorin,naringin,narirutin,hesperidin,neohesperidin,liquiritin and ammonium glycyrrhizinate in Xuefu Zhuyu Capsules.Method:Agilent TC-C18(4.6 mm×250 mm,5μm)was used as the chromatographic column,a mobile phase was acetonitrile-0.05%phosphoric acid in a gradient elution manner,the flow rate was 0.8 m L/min,the temperature of column was 30℃,and detection wavelength was set at220 nm.The relative correction factors(RCFs)of the other 6 index components were established and the content of each component was calculated by using paeoniflorin as an internal reference substance in this study.Results:Seven index compounds displayed good linear relationships within the range of 2.778-55.510μg/m L(r=0.9999),2.333-46.664μg/mL(r=0.9998),1.615-32.290μg/m L(r=0.9999),3.117-62.343μg/m L(r=0.9997),2.225-44.692μg/m L(r=0.9998),3.685~73.701μg/m L(r=0.9998),1.261-25.224μg/m L(r=0.9999),respectively.The average recovery rates were 99.88%(RSD=1.12%)(n=9),101.80%(RSD=1.10%)(n=9),105.07%(RSD=0.53%)(n=9),98.98%(RSD=1.21%)(n=9),101.22%(RSD=1.23%)(n=9),100.99%(RSD=1.31%)(n=9),98.79%(RSD=1.02%)(n=9),respectively.Conclusion:The QAMS method is accurate and reliable,and can be used for the quality control of Xuefu Zhuyu Capsule.
